we get a process route of Flavonoids Extraction by studying the detection methods, extraction, purification processes and antibacterial activity in vitro of flavonoids in Clinopodium gracile(Benth.)Matsumra. The main results are as follows:(1) UV-visible spectrophotometry was used to detect the concentration of the flavonoids,we detection methods for methodological study. The results obtained regression equation y=0.0115x-0.0026, correlation coefficient R2=0.9996, which indicated that its linear relationshipthe was good when the concentration of rutin was0-55μg/mL;the RSD of standards and the test solution stability test were1.54%and1.66%respectively; the RSD of precision and repeatability were0.93%and1.50%respectively; plus recovery test average recovery was98.42%, RSD was1.44%. the method is accurate and reliable.(2) The mathematical model were established according to2parameters:I:the response value was the extraction rate of total flavonoids which using PB screening and CCD design; II: the factor were ethanol concentration, liquid to solid ratio and ultrasonic time respectively. ANOVA showed that their goodness of fit is better. Fianally. an optimal ultrasonic extraction of total flavonoids route are as follows:ethanol concentration73%, extraction time32min, liquid to solid ratio of45mL/g, and the total flavonoids extraction rate is5.04%under this conditions.(3) The separation and purification of flavonoids using macroporous adsorption resinwas studied. Static adsorption, desorption tests showed that H-327B is an ideal adsorption resin as the separation and purification of flavonoids. Optimized process conditions:I:the sample concentration was9mg/mL and flow rate was3BV/h in the adsorption process; II:5BV ethanol (75%)was used to eluted in the elution process. the concentration of pure flavonoids extracted by macroporous adsorption resin is63.11%, which is5times than the Crude extracts(12.14%) of flavonoids in Clinopodium gracile(Benth.)Matsumra.(4) Antibacterial activity in vitro of flavonoids was studied. The flavonoids in the Clinopodium gracile(Benth.)Matsumra have good inhibitory effect on many kinds of bacteria, for example:its inhibition zone diameters flavonoids against Staphylococcus aureus, Escherichia coli, Streptococcus pneumoniae and copper fake Green Aeromonas were22.5mm,22.5mm,16.5mm and19.5mm respectively after being purified. Similrly, the MIC were1.5mg/mL,1.5mg/mL,0.5mg/mL and1.5mg/mL, and MBC were2.75mg/mL,1.75mg/mL,0.75mg/mL and3.25mg/mL. The results show that antibacterial activity of flavonoids improved greatly.
