Purpose: Tremella oligosaccharides of different molecule weight are prepared, weinvestigate their physiochemical and structural properties and examine the impact of differentmolecular weight Tremella oligosaccharides HK-2cells.Method: This experiment uses the acid hydrolysis, the hydrolyzate of to alcoholprecipitation Preparation of a mixture of polysaccharides and oligosaccharides, and themixture was separated by dialysis and717strong base anion exchange resin and732strongacid cation exchange resin mixed desalting column to give the oligosaccharide portion(TO-1).Sugar content (phenol-sulfuric acid method), determination of acidic sugar content (hydroxydiphenyl method), determination of protein content (BCA kit method) and PMP pre-columnderivatization, its physicochemical properties and monosaccharide composition analysis;using Sephadex G-15column separation on the TO-1, the HPLC method to detect theseparation results, the two parts of different molecular weight, measured both physical andchemical properties and monosaccharides composition analysis; Investigation of Tremellaoligosaccharide fragments of different molecular weights on the influence of HK-2cells;techniques used in conjunction with LC-MS analysis of samples TO-1, the composition of theanalysis wherein the oligosaccharide; derivatization on the TO-1, was prepared into thepartially methylated partially acetylated sugar alcohol derivative by GC-MS analysis of theconnection of its glycosidic bond.Result:The physicochemical properties of three different molecular weightoligosaccharides Tremella: TO-1total sugar content of about33.99%, the acidic sugar content of about7.05%, a protein content of about10.5%; TO-1-1of the neutral sugar content ofapproximately91.54%, acidic sugar content of about15.95%, a protein content ofapproximately10.91%; TO-1-2, the neutral sugar content of approximately45.04%, the acidicsugars content of about5.39%, a protein content of approximately11.87%. HPLC analysisshowed that the the white fungus oligosaccharides of three different molecular weightfragments by Man, Glu and GlcA; LC-MS results showed that the samples TO-1is a mixtureof oligosaccharides2-9sugar residues; GC-MS results showed that the TO-1is mainly in the1â†'3connected to the Man, there is a small amount of1â†'6and1â†'2connected,oligosaccharide larger molecular weight to1â†'3connected Man backbone, in the2womenwith side chains composed of Man,the terminal is mainly composed of Glu and Man,glucuronic acid in TO-1for the non-reducing terminal. The different molecular weight ofTremella fuciformis oligosaccharide promotes the proliferation of HK-2cells, and with theextension of time, the proliferation is more significant, the range of molecular weight had nodifference on HK-2cell proliferation in the230-1700D Tremella oligosaccharides.Conclusion:Be prepared by acid hydrolysis, the alcohol precipitation method of Tremellapartial acid hydrolyzate obtained after the separation and purification of Tremellaoligosaccharide, a preliminary analysis by LC-MS and GC-MS Tremella oligosaccharideschemical structure and different molecular weight was detected by MTT methodof whitefungus oligosaccharides HK-2cells, the study of the oligosaccharides of great significance.
