Angiotensin-converting enzyme (ACE) plays an important role in theregulation of blood pressure, and inhibit the activity of ACE can reduce bloodpressure. Foodborne ACE inhibitory peptides can reduce the side effects of theantihypertensive drugs of chemical synthesis, and its research and development areof scientific and pratical interest. In this study, choosing the spray-dried porcineplasma protein powder (PPSP) as object, pepsin as test enzyme, the effects ofpretreatment temperature, pretreatment time, the ratio of pepsin (E:S), enzymatictemperature, concentration of substrate protein, pH and enzymatic time on thedegree of hydrolysis (DH) and ACE inhibition rate of PPSP are studied by singlefactor and response surface tests, in order to optimize the conditions ofpretreatment and enzymatic hydrolysis, and established enzymatic quadraticregression model; and enzymatic degradation products were isolated and purifiedby macroporous adsorption resin (MAR), and the purification conditions wereoptimized. The results are described as follows:(1) The appropriate pretreatment conditions of getting ACE inhibitory peptides,suitable for pepsin enzymatic the PPSP, were as follows:80°C water bath andheating for20minutes.(2) The appropriate hydrolysis conditions: E:S3000U g-1, enzymatic temper-ature is37°C, protein concentration of the substrate is5%, pH is2.5, enzymatictime is3h. The order of combined effects of DH and ACE inhibition rate is pH> E:S> concentration of substrate protein> enzymatic temperature> enzymatic time,and the regression model is extremely significant (P<0.01), the regressioncoefficient R2is0.969.(3) The S-8MAR is suitable for the separation and purification of ACEinhibitory peptides of PPSP. pH of the sample solution for static adsorption is2.5,and the adsorption equilibrium time is3.5h. The eluent is70%ethanol solution,and the desorption equilibrium time is30min.(4) Under the conditions of25°C and pH of2.5, the concentration and flowrate of the sample solution, suitabled for dynamic adsorption of S-8is20mg/mLand1.5mL/min respectively.70%ethanol solution as the eluent, the flow ratesuitabled for dynamic desorption is1.0mL/min, and desorption time is80min.
