| Molecularly imprinted polymers have good development prospect in the separation of complex matrix with their high selectivity, stability and usability. In recent years, the application of biological samples was particularly concerned. Newly molecularly imprinted polymer was prepared and used as recognition material for selective recognition of LYS.The MWNTs-MIP film was prepared on the surface of the MWNTs using AAm as functional monomer, MBA as cross-linker, APS and TEMED as initiator under the optimized condition, which had selective recognition ability for LYS. The MWNTs-MIP film was characterized by TEM, FT-IR, TGA, and BET, respectively. The equilibrium adsorption experiments, dynamic adsorption experiments and the selectivity adsorption experiments were carried out to determine the binding capacity, the equilibrium time and specificity of the LYS-MIP film. The results showed adsorption capacity of the MWNTs-MIP film was20.89mg g-1at0.500mg mLL1, and adsorption capacity was15.37mg g"1for the MWNTs-NIP film.82.10%binging was obtained within30min and the adsorption equilibrium was achieved within300min. The MWNTs-MIP film had higher affinity capacity for LYS than that for the competition proteins, such as Cyt C, Mb, Hb and BSA. The relative selectivity coefficients for LYS/Cyt C, LYS/Mb, LYS/Hb, and LYS/BSA were1.30,1.30,3.12and2.82, respectively. Adsorption capacity of the MWNTs-MIP film was1.86times more than that of the MWNTs-NIP film.The MWNTs-MIP film was synthesized through preliminary work, then the MWNTs-MIFP film was prepared by adding FITC as fluorescent mark. The MWNTs-MIFP film was characterized by FT-IR. Subsequent adsorption of LYS caused quenching of fluorescence for the MWNTs-MIFP film. The fluorescence of MWNTs-MIFP film was stronger quenched by LYS than that of MWNTs-NIFP film, which indicated that the MWNTs-MIFP film could better recognize LYS. The imprinting factor was3.38.70.70%binging was obtained within60min and the adsorption equilibrium was achieved within300min. The MWNTs-MIFP film also showed selective detection for LYS among other proteins such as Cyt C, Hb and BSA due to the imprinted sites in the MWNTs-MIFP film.The imprinted polymer film had good adsorption property. This material has great development potential for detecing LYS, and may be used in the research field of solid-phase extraction, biosensor and bionic antibody and so on. |
