Abstract/Summary:
DNA is a kind of vital biologic large molecule, and it plays an important role in lifegenetic code of interpretation, transcription and copy. which is the main target molecule thatexists in antiviral、anticancer and antiviral drugs. It could be damaged under variousconditions, especially interact with some small molecules, so it could be applied in varioustype of diagnosis. The study on the interaction beween small molecules and DNA has becomean active research field of biochemistry and life science in recent years. Study of theinteraction between small molecules and ctDNA are helpful to elucidate the mechanism ofthese compounds of effects on the structure and physical properties of DNA and drug toxicityand pharmacokinetics. It is helpful to understand the modes of small molecules binding toDNA and DNA binding affinity to the design of new DNA-targeted drugs. The spectralstudies are important to understand the mechanism of the interaction of calf thymus DNAwith small molecular complexes. This thesis consists of four sections as follows:1. The important role of DNA in life activities, the fuction of DNA and the researchmethods of DNA have been introduced in this section.2. The interaction between naphthalene diimides derivatives and DNA in physiologicalbuffer (pH7.4) was investigated by UV-Vis spectroscopy,fluorescence spectroscopy,circulardichroism spectrum and DNA melting techniques.3. The interaction between novel Selective Estrogen Receptor Modulators (SERMS)coumarin derivatives and DNA in physiological buffer (pH7.4) was investigated by UV-Visspectroscopy,fluorescence spectroscopy,circular dichroism spectrum and DNA meltingtechniques.4. The interaction between novel antitumor coumarin derivatives and DNA inphysiological buffer was investigated by UV-Vis spectroscopy,fluorescence spectroscopy,circular dichroism spectrum and DNA melting techniques.
