Marine animals have a rich variety of biologically active substances and symbioticmicroorganisms, for scarce resources in today’s society, it has the potential to develop and useprospects. Symbiotic microorganisms and their metabolites produced by further studies toexplore the biological activity, to explore their potential in the food, pharmaceutical, aquacultureand other areas of open space for new development for the foundation.The protein, polysaccharides as well as bacterial of sea cucumber symbiotic Bacillussubtilis22respectively was researched in this study. They were determined the protein stability,polysaccharide fraction preliminary analysis, antifungal test, antioxidant test, in vitro anti-tumorexperiments, and polysaccharides on MMC-induced nuclear test civia.The basic physical and chemical properties of the test showed that protein22stability isgood for UV irradiation while the protein in trypsin, proteinase K, temperature and pH under lossof inhibitory activity. Protein22at-20℃,-4℃,20℃,40℃stored for6months antibacterialactivity did not change significantly. The polysaccharide IR and GC analysis showed that theabsorption peaks polysaccharide containing-OH,-CH2,-C=C-,-COOH, COC, C-OH,-O-SO3functional groups. Monosaccharides constituting of the polysaccharide22was relatively simple,mainly composed of arabinose, galactose and glucuronic acids.The antifungal test indicated that protein22could inhibite the growth of Trichotheciumroseum (Bull.) Link, Ralstonia solanacearum, Botrytis cinerea Pers, Gerlachia nivalis (Ces.atSacc.), Fusarium moniliforme, Fusarium oxysporum f. sp.Melonis, Corn alternaria solani,Fusarium oxysporum, Alternaria solani. Polysaccharide22could inhibite the growth of ninespecies of fungi, including Trichothecium roseum (Bull.) Link, Ralstonia solanacearum, Botrytiscinerea Pers, Gerlachia nivalis (Ces.at Sacc.), Fusarium moniliforme, Corn alternaria solani,Fusarium oxysporum as well. The bacterial cells of Bacillus subtilis on plant Botrytis cinerea testresult showed that was strongly inhibited against Botrytis cinerea. It gave tomato plants thatinoculated pathogen body of SOD, MDA, PAL, PPO, POD have some beneficial effects.The results of antioxidant protein and polysaccharide22tests showed that they respectivelysuperoxide anion and hydroxyl radicals have a strong clearance rate.1mg/mLl of protein22inthe superoxide anion radical and hydroxyl radical scavenging rates were45.40%and39.77%.When1mg/mL polysaccharide that superoxide anion radical and hydroxyl radical scavengingrates were34.66%and22.45%.The result of Anti-tumor experiment showed that protein and polysaccharide22had a stronginhibitory effect on neuroblastoma cells SH-SY5Y P7in vitro. When the protein22concentration was360μg/mL, the inhibition rate was28.88%.The polysaccharide22on neuroblastoma cells SH-SY5Y P7also had a certain extent, in the360μg/mL, the inhibition ratewas21.44%.The test results polysaccharide22on MMC-induced nucleation civia availability showdethat polysaccharide22on cell micronucleus formation significantly inhibited, when theconcentration of250μg/mL, inhibition rate up to53.91%.
