| The discharge of petroleum sludge is one of the obstacles to the sustainable development of petrochemical enterprises.Physical-chemical method is high cost and easy to cause secondary pollution.There are broad prospects for the use of microorganisms for harmless and resourceful sludge management,but it is only a concept that has been put forward,whose new concepts and technologies are not mature.Our research group got some microorganisms with rapid metabolism of hydrocarbons.This paper explores the idea and feasibility of using these microorganisms to make the oil sludge harmless and recyclable.The study is of great significance for us to solve the environmental problems caused by oil sludge in petrochemical enterprises.In this paper,we use thin layer chromatography(TLC),Fourier Transform Infrared Spectrometer(FT-IR)and high performance liquid chromatography-time-of-flight mass spectrometry(HPLC-TOF-MS)for the analysis of detection.and study the characteristics of Pseudomonas aeruginosa NY3 and its symbiotic microorganisms in the treatment of oil sludge and its characteristics of biosurfactant.Get the following results:(1)NY3 strain and its three symbiotic bacteria in a single bacteria or mutual compound conditions,can grow in the high concentration of oil sludge(converted into net oil content 15 g/L).Among them,the single FF bacteria and "NY3 bacteria + FF bacteria" compound on the sludge in the main pollutants are the best degradation of alkanes.The total degradation rate of the main pollutants in the main pollutants are 90.98% and 82.29% for 4 days.In the metabolism and its rapid removal of oil,the bacteria can be converted into oil up to 7.54 g/L extracellular macromolecules.(2)FF bacteria not only can quickly degrade hydrocarbons,but also is anti-high concentration of oil(equivalent to the net oil content of 10 g/L).The oil sludge whose concentration is 2 g/L can be almost completely degraded with FFbacteria in 2 days,and it can remove 80% of the pollutants in oil sludge whose concentration in 10 g/L.Besides the most hydrocarbons convert into lipid high-energy substances accumulated in the cells.Especially,when the oil content increases in degradation system,the phenomenon of lipid accumulation is more obvious(3)FF bacteria can produce biosurfactants during the degradation of oil sludge.The stability of emulsions formed by the biosurfactants will be better at the appropriate concentration.which can improve the hydrophobicity of the FF cells and accelerate the degradation of hydrocarbons by accelerating the mass transfer rate of pollutants.(4)It is approved that the biosurfactant produced by FF strain is a mixture of trehalose ester,and the main components are trehalose diester and triester.The polarity of the biosurfactant is similar to that of rhamnose monolayer surfactants produced by NY3 bacteria,which is less than that of rhamnose(5)The co-carbon source fermentation which has FF bacteria to oleic acid + hexadecane(as a representative of hydrocarbon contaminants)can secrete more trehalose lipid surfactants.Under the optimized conditions of the content of carbon,nitrogen and phosphorus in the medium,and chloroform: methanol =1:2 as the extractant,the yield is up to 4.588 g/L. |
