Milk powder and chicken is the important animal-derived food in people life andconcerned as food safety events in recent years. Hormones as endocrine disruptors affect onadult and adolescent health. So it is important to qualitatively and quantitatively analyze andensure food safety.Two kind of fast, efficient, accurate analysis methods were established to detect5sexyhormones in milk powder and chicken. The conditions for research were the samplepreparation, HPLC and GC-MS. The main investigation factors included extraction solvent,degreasing solvent, solid phase extraction conditions and derivatization conditions; HPLCdetection wavelength, column temperature, HPLC column type, HPLC mobile phasecomposition; GC-MS temperature program and the injection temperature. The bestexperimental conditions were15mL extraction solvent for acetonitrile,10mL degreasingsolvent for n-hexane, the solid phase extraction column for HLB, eluting solvent for methanol,derivatization reagent for50μL pentafluoropropionic anhydride, derivatization temperature of30℃; detection wavelength for230nm, column temperature for30℃,the mobile phasecomposition for acetonitrile: ammonium acetate=80:20; the temperature program is70℃(1min)â†'30℃/minâ†'180℃(1min)â†'4℃/minâ†'240℃(2min) and injection temperature is230℃. The GC-MS and HPLC method are accurate, reliable and five kinds of hormones ina wide range of concentration (0.1-50μg/L;1-40μg/mL) have a good linear relationship. Thelimits ranges of detection are0.14-0.50μg/Kg and1.3-3.9μg/Kg respectively. The recoveriesare90.0~106.9%and90.9~95.7%respectively. Meanwhile5kinds of sexy hormones in thechicken and milk powder of three batches were not detected using the option method.
