Peptide antibiotics and alicylanilides are broad-spectrum antimicrobial and vermifugeveterinary medicine and their residue in foodstuffs of animal origin have certain toxical effecton human health. Pepele pay attention to the food safety issue produced by above mentionedelements more and more. The representational animal tissues were selected as study targets inthis research, and have a study on the determination method of closantel and rafoxanideresidue and bacitracin, polymyxin B, colistin, virginiamycin residue in foodstuffs of animalorigin by high performance liquid chromatography(HPLC) and high performance liquidchromatography-tandem mass spectrometry(HPLC-MS/MS) technology respectively. Theresult show that the method could satisfy the relative international detection demand perfectly.The research consists of two parts.Part one, animal tissues, such as muscle, liver and kidney were selected as study targets,and established a method to detect the closantel and rafoxanide residues of them by Highperformance liquid chromatography. The samples were extracted with acetonitrile, isolated bya5C18Liquid Chromatographic column(250mm×4.6mm,5μm)and detedted at wavelengthof282nm by UltraViolet detector. The research results indicatied that the averagerecoveries closantel and rafoxanide ranged from71.36%~92.23%and70.86%~95.66%and the relative standard deviation(RSD)of them were2.05%~7.06%and3.02%~7.03%respectively. And the lowest limit of detection (LOD) are10~200μg/kg for Closantel andRafoxanide. This study method is simple, accurate and sensitive and is suitable for fastscreening and detecting the residues of closantel and rafoxanide in animal tissues.Part two, animal tissues, such as muscle, liver and kidney and milk were selected asstudy targets, and established a method to simultaneous detect the Bacitracin, polymyxin B,Colistin, virginiamycin residues of them by liquid chromatography mass spectrometry. Thesamples were extracted with methanol–water containing0.1%formic acid, degreased byhexane and cleaned up by solid phase extraction (SPE) column. The elute was separated byliquid chromatography and determined with the multiple reaction monitoring mode by mass spectometry. The research results indicatied that the average recoveries Bacitracin,polymyxin, polymyxin B, Colistin A, Colistin B, virginiamycin ranged from88.54%~99.84%, the relative standard deviation(RSD)of them was1.91%~9.81%. The method is simple,rapid, accurate and suitable for simultaneous determination of peptides antibiotics in differentanimal tissues.
