Syrmaticus humiae is classified as the animal of Phasianidae, its scarcity, distribution range is small. Belong to national I level to protect animals. In1995it was listed in the convention on international trade in endangered species of wild fauna and flora I the appendix. The world conservation union (IUCN) red list of endangered species as endangered species in2000.Understand the diffusion pattern of Syrmaticus humiae and habitat selection, is for the Syrmaticus humiae the prerequisite for effective protection, and the existence of the genetic diversity of species can maintain the evolution of a species. Based on molecular level and the study of Syrmaticus humiae are less, this study adopts the microsatellite ISSR molecular markers for Syrmaticus humiae genetic relationship and genetic diversity analysis, for Syrmaticus humiae molecular basis for the study of the diffusion pattern, enable us to better protect it, to avoid this type of endangered pheasant extinction.This study using microsatellite ISSR technique to analyze, and primers is use Canada’s Columbia University published100ISSR primers for Syrmaticus humiae amplification, found that there are7primers amplification bands with amplification repeatability is good, with clear and strong specificity, quantity, and the sample concentration, primer concentration of DNA and each primer PCR reaction gradient the annealing temperature and cycle times with screening to establish the experimental optimal ISSR-PCR amplification system, improve the stability of the ISSR-PCR.Research results show that with good polymorphism and repeatability of article7primers, PCR amplification produced a total of100belt of amplification, the average each primer amplification of article14.3, of which88are polymorphic, polymorphism rate reached88%, showing rich polymorphism, the number of alleles detected Na=1.8800, the effective number of alleles Ne=1.4042, Nei’s genetic diversity index (He) is0.2514, Shannon’s information diversity index (I) is0.3916, total gene diversity (Ht) is0.2533, the genetic diversity within population (Hs) is0.2341, Syrmaticus humiae have higher genetic diversity, and studies have shown that Syrmaticus humiae captive population genetic diversity level significantly lower than the level of population genetic diversity of wild, genetic differentiation (GST) was0.0757, shows that Syrmaticus humiae genetic variation of92.43%is from within a population, only7.57%of the genetic variation between populations, most of the genetic variation come from within a population, gene flow (Nm) is6.1022, Syrmaticus humiae has certain gene flow between groups.By using Ntsys software to calculate the genetic similarity coefficient between individuals, Syrmaticus humiae25clustering analysis, the similarity coefficient of0.49-0.86, among them2individual and17individual genetic similarity coefficient is the highest, it is0.8600, but9individuals and21individual genetic similarity coefficient is the lowest, only0.4900,25individuals can be divided into four groups.
