Interferon is a kind of cell factor produced by monocytes and lymphocytes, which can resist viruses, affect cell growth and differentiation,regulate immune function and other biological activity in the same kind of cells.Interferon-inducible p200protein family members are highly conserved, consists of6murine proteins including p202a, p202b, p203, p204, p205and p210as well as4human homologues including IFI16, MNDA, AIM2and IFIX. The p200protein family members have at least one HIN domain at C-terminus and one Pyrin domain at N-terminus which are related to cell growth, existence and differentiation. C-terminus of the family members have one or more specific200amino-acid domaims.p204which has640amino acids is the homologous protein of IFI16, which is a member of human p200protein family. The apparent molecular weight of p204is72kDa. p204contains a nuclear localization signal and nuclear export signal at its N-terminal, and two conserved200-aa segments at its C-terminal which pRb motif binds and locates on. p204cooperates with tumor-suppress protein pRb to regulate cell differentiation as well as other transcription factors. p204can also inhibit cell proliferation and work as negative feedback suppressor of oncogenic protein K-ras to participate in the physiological regulation of organisms. Therefore, p204may play an important role in tumor immunity.p200family proteins are widely involved in regulating cell proliferation, differentiation, senescence and apoptosis by combined with a variety of cytokines or transcription factors and reactions. Its abnormal expression is associated with many diseases, such as autoimmune diseases, inflammation, and tumor related diseases. p200family proteins play an important role in some fields such as autoimmune responses, antiviral and anticancer. Study on p200protein family gene expression pattern will contributes to further research on its function and clinical application.This experiment used C57mice as the model animal by RT-PCR to test the expression of gene p202, p204, p205and p210in different stages of mice early embryonic development and different tissues of adult mice, and analysed their expression pattern.The result showed that the expression of p204and p210reached the maximum value at the ninth day of embryonic development But both of them kept decreasing as the development of embryo, and went up again till the19th day. The expression pattern of gene p204and p210are almost the same. The expression level of p202, p205and p210were highest in spleen tissue, then in thymus tissue, and were lowest in heart tissue. The expression of p202in heart and spleen in old group was higher than in young group. The expression of p205of old group in thymus and heart were higher than in young group, and in spleen is reverse.Expressions of p200protein family members in heart tissue were roughly higher in old group than in young group, and so in immune tissue. As for the abnormal expression of p210in spleen, it might be related to the association of p210with inflammation caused by the external environment disturbance in mice.The results indicated that p200protein family had similar expression pattern. No matter in young stage and old stage, expression of the family members were highest in hemopoietic tissue spleen and lowest in heart. This can explain why p200protein family members were firstly found in hemopoietic tissue and later in other tissues. High expressions of p200protein family members in immune tissues showed that this family members may highly relate to autoimmunity, but their expression altered in different ages. We speculated that the expression might associate with the development of immune organs in mice.
