We chose the pit mud samples as our Study object using metagenome and454pyrosequencing,combine with bioinformatics to analyzed the distribution pattern of microbial diversity in fermentatian of liquor.The results are as follows:1.Bands from No.10to No.18were showed clearly in all pit mud samples.The pronucleus microorganism’s community of the same pit during different fermentation period were similar.Besides,the mean diversity index of all samples kept between2.20and2.88.With the increasing of pit age,the diversity index of samples from the same spots generally showed increasing trend.The similarity index of PCR-SSCP pattern of samples from different spots in the same pit was higher, between0.424and0.600.While the similarity index of samples from different pit ages was lower,between0.261and0.700.2.Analysis of pronucleud microorganism’s community using454 pyrosequencing.Belonging to18phylum,23genus and species of the total bacterial OUT are1183in all pit mud samples.In distribution of time,Similar succession in pronucleud microorgrenism’s community.In distribution of Spatial,The abundance have slightly different in advantage flora.Firmicutes have a dominant position throughout the fermentation.In the early of the fermentation, Bacilli, Lactobacillales, Lactobacillaceae, Lactobacillus have a dominant position. Clostridia, Bacteroidia, Bacteroidales, Bacteroidaceae and Bacteroides have higher abundance in initial fermentation period.In the late of the fermentation,Clostridia, Clostridiales,Ruminococcaceae have advantages,and sequencing of "Other" accounted for a large proportion.3.Bands from No.3to No.6were showed clearly in all pit mud samples.The eukaryotic microorganism’s community of the same pit during different fermentation period were similar.Besides,the mean diversity index of all samples kept between1.10and1.79.With the increasing of pit age,the diversity index of samples from the same spots generally showed increasing trend.The similarity index of PCR-SSCP pattern of samples from different spots in the same pit was between0.44and0.75.While the similarity index of samples from different pit ages between0.22and0.91.4.Analysis of eukaryotic microorganism’s community using454pyrosequencing.Belonging to5phylum,9class,10order,13family,18 genus,and species of the total eukaryotic OUT are64in all pit mud samples.In distribution of time,Fungal’s community composition is relatively simple.In distribution of spatial,the abundance have slightly different in advantage flora.Five classification level Between phylum,class,order,family,genus,strain of Opisthokonta、Nucletmycea、 Fungi、Dikarya、Ascomycota have dominant position,abundances of up to94%.
