Calcium signals involved in various responses to abiotic and biotic stimuli, including light, high and low temperature, touch, salt and drought, osmotic stress, plant hormones, pathogen elicitor, and nodulation factorsand then may transmit signals downstream to basic defense reaction.Signal information would be encoded by a specific Ca2+signature that is defined by precise control of spatial, temporal and concentration parameters of alterations in cytosolic Ca2+concentration Non-selective ion channels (NSCCs) are common ion channels in plant membranes, and are selectively permeable for cations over anions. AtGLRs are non-selective ion channels that have attracted considerable attention as important regulators of Ca2+influx. In Arabidopsis,20genes encode for AtGLRs that are differentially activated by Glu and Gly, as well as by other amino acids, and mediate an increase of cytosolic Ca2+.Recent studies show that AtGLR3.3showed enhanced susceptibility to the bacterial pathogen Pseudomonas syringae pv.tomato DC3000.We cloned AtGLR2.9promoter into the binary expression vector pORE R1and got Promoter AtGLR2.9::GUS. Through Agrobacterium infection wild-type Arabidopsis and multi-generation screening, we got Arabidopsis T3genetically modified seeds stably expressing Promoter AtGLR2.9::GUS. Then using GUS gene tissue-specific expression activity to locate AtGLR2.9in leaf vein and root tip of Arabidopsis. In addition, by using MQA culture medium in various ions、amino acids and hormones condition, we found the number of lateral root of Col-0are strongly inhibited in500μM Cys MQA culture medium. This study will expand the understanding of Arabidopsis AtGLR gene family and provide the basis to explan the mechanism of calcium channels, to clarify the role of calcium signaling in organism.
