Calcium is the essential mineral and nutrient element for plant growth and development, as the important second messenger translates the outside stimulation into calcium signal and downstream signal response, and produced physiological and biochemical reactions inside the cell to respond and adapt to the external environmental stimuli. Calcium ion involved in various responses to abiotic and biotic stimuli, including temperature, light, drought, salinity, plant hormone and pathogen infection. Cyclic Nucleotide-Gated Channels (CNGCs) are one kind of non-selective cation channels in the animal and plant. There are 20 members in Arabidopsis CNGCs familyl. CNGCs in plants play important role in physiological function, such as ion transport, growth and development, pollen tube elongation, pathogen defense response, gravitropism, heat resistance, adversity stress, and so on.We performed homozygous verification of the CNGCs family T-DNA mutants and then we used the homozygote mutants to phenotypic analysis, after 25 days we found the mutants as compared with Col in 1mM Ca2+ medium have no significant difference, but in 20mM Ca2+ medium the mutants cngc2-2 and cngc4-2 and their point mutants cngc2-1 (dnd1) and cngc4-1 (dnd2) showed drastically stunted growth.We with atomic absorption spectrometry found that in 1/2 MS+20 mM CaCl2 medium the content of Calcium in cngc2-land cngc4-1 was very significant less than Col.By using Aequorin to monitored the dynamic change of cytosolic Ca2+ concentration ([Ca2+]cyt), we used composition transgenic plants Col::AEQ and dnd1::AEQ to compare the different [Ca2+]cyt between cngc2-1 and Col. We found in 1mM Ca2+ treatment there are no significant different between the [Ca2+]cyt transient rise in cngc2-1 and Col.This study will expand the understanding of Arabidopsis CNGC gene family and provide the basis to explain the mechanism of CNGC channels, to clarify the role of CNGC channels in organism.
