With the human activities and natural factors and the rapiddevelopment of industrialization,the environment is beingdestroyed. The UV-B radiation which reached the surface of theearth would increase with air pollution caused by the ozone hole.The enhanced UV-B radiation caused great influences in theplant growth development, metabolism, the Earth’s ecosystemsand humans. And UVR8identified recently acted a UV receptorsis vital.UVR8protein which identified in the Arabidopsis cells isUV-B light receptor. In this study, we treated Arabidopsis andwheat with set of four treatment groups,which were controlgroup, the UV-B radiation treatments, UV-B radiation and He-Nelaser irradiation combined treatment group, and the He-Ne laserradiation respectively.Then researched the seedling growth andphysiological indicators on Arabidopsis with different intensityand times of UV-B radiation from morphological,physiological and biochemical; and preliminary screeninged the parametersof UV-B radiation;and studied the effects of UVR8in Arabidopsiswith He-Ne laser and UV-B radiation treatments on the level ofgene transcription;and detected UVR8in wheat by using PCRamplification technology,then sequenced and done sequenceanalysis comparison. The results were as follows:1.The growth and development of Arabidopsis was inhibitedwith the increasing of UV-B radiation intensity,while waspromoted with the low-intensity UV-B radiation to some extent.2.Under short period of UV-B radiation, the root length wereelongated, chlorophyll content and soluble protein content wereincreased in Arabidopsis leaf; but prolonged period of UV-Bradiation inhibited the elongation of root length, caused leafchlorophyll content, soluble protein content and chlorophyllfluorescence characteristics such as Fv/Fm and qP beendecreased, promoted SOD activity, MDA concentration, CATactivity and qN been increased together, and also graduallydecreased or elevated over time.3.When the radiation power was16.67μW·cm-2, the optimalperiod of time of UV-B radiation was1.5h.4.Tested UVR8expression of Arabidopsis with treatmentsby using quantitative PCR,the results indicated that the UVR8 relative expression followed by the control groups, UV-Bradiation treatments groups, He-Ne laser irradiation treatmentgroups and UV-B and He-Ne laser irradiation combinedtreatments groups.5.Tested UVR8expression of wheat by using PCRamplification,and then sequenced the resulting amplifiedsequences,and the results indicated that the amplifiedsequences had high sequence homology up to85%with UVR8of millet cells,suggested that wheat contains UVR8.
