ABA is theimportant hormones in regulation of plant growth and development and in response tobiotic and abiotic stress, the molecular mechanism of ABA signal transductionpathway for the promotion of the development of modern agricultural production hasgreat significance.This study using the Infusion technology successfully cloned DDB1A,constructed to the plant expression vector pEGAD,by using the method ofagrobacterium mediated infection in arabidopsis thaliana, use Basta screening andLuminometer instrument instantaneous detection, received the DDB1A overexpressionof transgenic plants, and then through numerous homozygous, received T3generationof transgenic plants.by western blot test for DDB1A T3generation of transgenic plants, that DDB1Ahave been integrated into arabidopsis thaliana genome, and successfully expressed.Using the method of three primers to identify the homozygous DDB1A mutant,identified SALK038757C is homozygous mutant.DDB1A (gene locus AT4G05420) is a kind of damage of DNA binding protein, itcan with CUL4-RING ubiquitin ligase form a complex, it involved in the process ofplant hormone signal transduction, In this experiment DDB1A is regulated genes byABA. then experiment phenotype showed DDB1A sensitivity to ABA, but mutantddb1a is not sensitive to ABA, Description DDB1A is ABA’s response genes, thepromoter sequence analysis also showed that ABA in DDB1A promoter region there isa response of components, real-time quantitative rt-pcr also indicated that DDB1A in various organizations have expressed, the expression of flower is the most, analysismay participate in floral organ development, and the response induced by ABA intransduction level.
