Ethylene(ET), as a gas hormone in plants, played important roles in mediating the growth and development, and responsing to biotic and abiotic stresses in rice. The TPR motif is one of many repeat motifs that form structural domains in proteins that can act as interactional scaffolds participating in phytohormone signaling responses. Our project group found out that OsTPR1,a member of TPR proteins in rice, interacted with OsERS1, one of ethylene receptors by Y2H (Yeast-two-hybird) system. Further more, we also identified that the transcription level of OsTPR1and OsERS1in14days age of wild-type rice seedlings were subsequently elevated with the treatment of ethylene in dark through real-time PCR assay, and implied that both OsTPR1and OsERS1participated in ethylene signaling transduction pathway in rice. Based on these evidences, we confirmed the characters of interaction between OsTPRl and OsERSl through GFP subcellular localization, Y2H, GST pull-down and BiFC(Bimolecular Fluorescence Complementation) further. GFP subcellular localization indicated that OsTPR1and OsERS1may interact in onion cell, and we also identified that OsTPR1can interact with OsERS1in cytoplasma through Y2H and BiFC. Besides, we got the inducible proteins including GST-TPR1, His-ERS1, His-GAF-HisKA, His-HisKA-HATPase_c and His-GAF-HisKA-HATPase_c. Meanwhile, we also acquired the transgenic rice seedlings with overexpression or low expression of OsTPR1(K281:OE-TPR1; K279:RNAi-TPR1). As a result, we further proved that the interaction between OsTPRl and OsERS1, and infered that OsTPR1participated in ethylene signaling transduction pathway associated with OsERS1. Besides, the mechanism of OsTPR1involved in ethylene signaling transduction pathway would be explored with the successful breeding of transgenetic meterials.
