Glycine betaine is an osmoprotectant, it plays a key role in environmental osmotic stress.Phosphoethanolamine N-methyltransferase(PEAMT)is the key enzyme in the synthesis ofcholine. Choline is the precursor in the synthesis of glycine betaine(GB). This study analyzethe expression of Phosphoethanolamine N-methyltransferase gene from Suaeda liaotungensisK. and evaluate the function of SlPEAMT promoter.Specific work and results are as follows:We use Semi-quantitative PCR and real-time quantitative PCR techonlogy to analysisSlPEAMT gene expression characteristics and GUS gene which from transgenic pP-GUStobacco expression characteristics under stress treatment (salt, cold, drought, ABA).Results showed that, under normal condition the expression of SlPEAMT gene showed inall organs, with the highest expression in leaves, lower in stems, the lowest in roots. Whileunder salt stress, cold stress and drought stress, SlPEAMT gene has a response in all organs,so we conclude SlPEAMT gene is a stress-induced gene. SlPEAMT gene also has a responseto ABA treatment, it maybe an ABA dependent gene.For pP-GUS tobacco, under normal condition, GUS gene expresses in all organs,with thehighest expression in leaves, lower in stems, the lowest in roots. While under salt stress, coldstress, drought stress and ABA stress, GUS gene has a response in all organs, so we concludethat GUS gene expression is induced by stress.The expression of GUS gene in transgenic pP-GUS tobacco showed the same tendencywith SlPEAMT gene in Suaeda liaotungensis, so we have reasons to believe that SlPEAMTpromoter is a stress-induced promoter.
