NAC is a kind of plant-specific transcription factors that regulate growth and development and the response to abiotic stress.Transcription factors and their corresponding cis-regulatory sequences act as molecular switches for gene expression, regulating their temporal and spatial expression.Suaeda liaotungensis K. is a indicator plant of saline-alkali land in liaoning coastal area, they can live in the nature high salinity area, this indicate that Suaeda liaotungensis K. is valueable and contains a large number of genes related to resistance to salt stress.High salinity belongs to a kind of abiotic stress, so Suaeda liaotungensis K. may also have certain resistance to other abiotic stress.Through the study of genes and their mechanism of resistance to abiotic stress can enhance the abiotic stress resistance of other economic crops and provide more theoretical basis and genetic source.This study identified an EST from the transcriptome sequencing database of S. liaotungensis K., and named it with SlNAC8. By PCR technology,obtain a correct 750 bp sequence with complete open reading frame, the length of the ORF is 576 bp, encoding 191 amino acids. Analysis and predict SlNAC8 biology function by phylogenetic tree analysis and protein alignment comparison.By using qRT-PCR method,we analysis the tissue characteristics of SlNAC8 and the expression of SlNAC8 under salt, drought, low temperature and ABA stress. SlNAC8 gene expressed different patterns in different tissues under normal condition. Leaves was the highest, lower in stems, the lowest in roots. Under different abiotic stress,the expression of SlNAC8 showed a trend of increase and the highest expression appear at different times.And in salt,low temperature and ABA stress, the expression of SlNAC8 shows an significant trend. SlNAC8 genes response to salt, low temperature, ABA stress.The distribution of pEGAD-GFP-SlNAC8 was transfered into onion epidermal cells by using gene gun. Subcellular location assay showed that GFP-SlNAC8 protein was targeted to the nucleus. The transcription activation assay in yeast demonstrated that SlNAC8 and its C-terminal had the transcription activation activity. Subcellular localization results and transcription activation activity analysis proved that SlNAC8 were transcription factors.The distribution of pBI121-SlNAC8 was transfered into Arabidopsis by infloresence infection method. We obtained SlNAC8 transgenic Arabidopsis. Through screening,we obtained the homozygous diploid and analysis the resistance under three kinds of abiotic stress.Transgenic experiment indicated that SlNAC8 was an excellent gene to salt resistance, ABA resistant and cold resistant, and the resistance to drought is also particularly significant. Therefore, the plant-specific SlNAC8 gene plays an important role in the process of resisting abiotic stress and it is a very valuable transcription factor of resistance to abiotic stress.
