Collagen is a kind of white opacity native celloglobulin without branched chain, and it isresided in skin, skeleton, teeth, tendon and ligaments, blood vessel generally. Collagen is oneof the most abundant proteins in the mammal body, the main function of which is to keep theappearance construction of shin and organs, support organs and protect the body. The maincomposition of fish bone and connective tissues is collagens. In recent years, with thedevelopment of the methods of extracting collagen protein and deep research of its structureand properties, the functional properties of collagen and its peptide are widely realized, theresearch and application of collagen have become a hotspot in biological and medical industry,food industry, cosmetics industry, etc.In this paper, first,the cod skin was used as raw material to extract collagen; second,Czapek’s medium、collagen medium and gelation selection medium were used to screen outthe bacterial strain contain collagenase’s activity from the mud near the fish market and beach,and the enzyme activity was determined; then, put extracted collagen and collagenase to carryout enzyme reaction in order to determine the optimum condition; finally the collagen activepeptide was analyzed by SDS-PAGE electrophoresis method and the hydroscopicity、oilabsorption、foamability and its stability of short peptide and collagen were also researched.The result showed that the strain screened out from two kinds of mud, was identified asBacillus subtilis initially through the Physiology-biochemistry experiments, the enzymeactivity of collagenase was44.982U/mL. When the collagen and enzyme were mixed in theproportion of2:1,reacted at37℃for10h, and the collagen of100ku was hydrolyzed intocollagen peptide of8ku mostly. Compared the collagen with the collagen peptide, it wasconcluded that hygroscopicity of collagen increased obviously with reduction of molecularweight. After hydrolysis, it’s oil absorption also reduced. Foaming property of collagendropped with the hydrolyzate’s molecular weight.
