The silkworm has been used as an edible insect and is rich in resources in China, and itsrelatively low utilization. This research could improve the application value of silkworm byextracting the silkworm pupal oil, and to promote the economic benefits of the silkworm puparesources. Based on this, preparing protein oil emulsion medicine with silkworm pupa oil forresearching the medicine oral administration absorption, we discovered that the proteinmedicine emulsion prepared with silkworm pupa oil could promote oral absorption.In this research, single factor experiments and orthogonal array design were adopted tooptimize the parameters of silkworm pupal oil extraction. The effects of independentparameters, including temperature, extraction time, solvent volume and oscillation rate on theyield of oil were studied and the optimal extract condition was obtained. The statistical analysisshowed that the solvent volume was the most significant variable followed by extraction time,oscillation rate and temperature. Under this condition, the extracting rate of pupal oil wasmeasured about28.13%.Fatty acid composition of pupal oil obtained was analyzed by GC-MS. The silkwormpupae oil was rich in unsaturated fatty acids which accounted for more than68.34%of the totalfatty acids. The main unsaturated fatty acids are oleic acid and α-linolenic acid, and the mainsaturated acid in the silkworm pupal oil palmitic acid and stearic acid.Presently, protein drugs are mostly given to patients by injection, which caused greatsuffering to the patient and accompanied with a variety of adverse reactions. And the oraladministration may be solved these problems. However, the protein drugs would be enzymaticdegraded within the gastrointestinal and lose activity after oral administration. It causes oraladministration absorption rate to be extremely low of protein drugs. Taking the silkworm pupaoil as an oil phase, Span-80and Tween-80as surfactant, silkworm pupaoil-water-Span-80-Tween-80are determined in the process of W/O/W composite emulsionpreparation. Through single papameter and orthogonal experiment design analysis, the optimalpreparation method is determined.The composite emulsion can protect protein drugs from being degraded by embed in thedomestic waters. Releasing drugs lowly through oil phase to achieve the aim of sustainedrelease. Mice were orally administered with silkworm pupa oil W/O/W composite emulsion which was loaded rhGM-CSF. The results showed that the rhGM-CSF concentration in theserum of mice treated with W/O/W composite emulsion loaded rhGM-CSF were249.04pg/mLand238.04pg/mL at40min and60min after administration respectively. It showed that theW/O/W composite emulsion could protect rhGM-CSF not to be destroyed, and then increasethe absorption rate.
