Styrax dasyanthus Perk. is one of the most widely distributed in Styrax, has high ornamentalvalue, and is a valuable ornamental flowers, fruit, medicinal and oil resources aromatic plants,also is the value of comprehensive development and utilization of a large and broad applicationprospects, but current related research had not been reported. In order to develop and utilize it, thepropagation of seed and hardwood cutting and its rooting mechanism, as well as genetic diversityanalysis by microsatellite of this species had been studied in the dissertations. The main resultswere as follows:1.S. dasyanthus seed belongs to comprehensive dormancy, the reason of seed dormancyincludes episperm barrier, physiological dormancy, deficient of germination hormones andgermination inhibitors existing in both embryo and endosperm; and the highest of inhibitoryactivity was embryo, endosperm was higher than episperm, and physiological dormancy maybethe primary limit factors. GA3treatments were used to improve the germination rate of the seeds,and the embryo dormancy can be relieved after GA3treatment and stratification at lowtemperature, and one of the effective ways was by1200mg/L GA3treatments24h for30days, but120d was better, germination rate was up to44%.2. Top, middle and basal parts of cutting existed some materials in the phloem, whichinhibited seed germination and growth of Chinese cabbage in different extent, and the inhibitionbetween different parts of branches also have obviously differences, the contents of the materialsrelatively higher in the basal part cutting, and it influenced and inhibited rooting of cuttings, sothe cuttings should be selected from middle to upper of the branch for propagation material.3. S. dasyanthus is the plant of the surface root type by preliminary judgment. Four kinds ofgrowth regulators could significantly improve the cuttings rooting index; the most significanteffect on hardwood cutting by the treatment of500mg/L IBA30min, the rooting rate was thehighest, and rooting rate, root number and the longest root length were62.25%,7.53and93.29mm, respectively. IAA, NAA treatment followed by.4. Nutrient content and antioxidase activity in hardwood cuttings of S. dasyanthus wasclosely related to rooting, and the rooting rates often associated with soluble sugar, solubleprotein, POD, SOD and PPO activity, that was positive correlation, but was negatively correlatedwith IAAO activity.5.Eighty-seven samples of six natural S. dasyanthus populations were taken, and a total of43alleles were detected, with a mean of10.63per primer by four pairs of microsatellite primers. Themean percentage of the polymorphic loci of all the populations (P) was100%. The mean ofobserved heterozygosity and expected heterozygosity were0.659and0.811, respectively. According to coefficient of gene differentiation (Fst=0.118), the gene flow was higher (Nm=2.050),and the percentage variation among the populations was11.8%, while the percentage variationwithin the populations was88.2%, consistent with the conclusion of phenotypic variationresearch roughly, illustrating that the genetic differentiation of S. dasyanthus was quite low, andinbreeding within population (Fis=0.192). Consequently, the suggesting that was a high geneticdiversity of natural S. dasyanthus populations.
