Three stain of fungi, two of which are endophytic fungi Pestalotiopsis, one is phytopathogenic fungi, are studied in this paper for their metabolites chemical constituents and bioactivity. Three strains are fermented in large scales. The products after fermentation were extracted and purified by means of silica gel column chromatography, gel filtration chromatography, preparative thin-layer chromatography, HPLC, and so on. Structures of the compounds were identified by extensive spectroscopic analysis, such as1H-NMR,13C-NMR, HMBC, HSQC,1H-1H COSY, NOESY, MS, IR. Some of the compounds were tested for the antitumor activity and antioxidant activity. Twenty-nine compound were separated and identified in all, in which there are four novel compound.Pestalotiopsis calabae is endophytic fungus from Annona glabra Linn. Twelve compounds were separated from this strain after liquid fermentation and there are four novel compounds in the world. The structures of compounds were ergosta-4,6,8(14),22-tetraen-3-one(2-1-1),(+)-(4S,8S)-foedanolide (2-1-2),(+)-(4R,8R)-foedanolide(2-1-3), p-hydroxybenzoic acid (2-1-4),1’,2’-epoxy-3’,4’-didehydropenicillide (2-1-5), succinic acid (2-1-6), pestalotin (2-1-7),4-(2-hydroxypentyl)-4-methoxy-3,7-dioxabicyclo [4.1.0] heptan-2-one(2-1-8), hydroxypestalotin(2-1-9),2,6-bis [(1R)-1-hydroxyisobutyl]-1H,5Hpyrrolo [2,3-b] indole-4,8-dione(2-1-10)6-(1,4-dihydroxypentyl)-4-methoxy-5,6-dihydro-2H-pyran-2-one(2-1-11),6-(1,3-dihydroxypropyl)-4-methoxy-5,6-dihydro-2H-pyran-2-one(2-1-12). Compounds2-1-11and2-1-12were novel compounds.Pestalotiopsis calvispora is endophytic fungus from Psidium guajava L.. Eleven compounds containing two novel compounds were separated from this strain. The structures were ergosta-7,22-dien-3β,5α,6α-triol (cerevisterol)(2-2-10),3β-hydroxy-ergot steroid-5,7,22-triene (2-2-1),3β-hydroxy-5α,8α-ergot peroxide steroid-6,22-diene (2-2-8),4-hydroxy phenyl ethyl acetate (2-2-2),4-hydroxy benzaldehyde (2-2-3),2-hydroxyphenylethanol (2-2-6),4-hydroxyphenethyl alcohol (2-2-7),4-hydroxybenzyl alcohol (2-2-9),4-hydroxy-4-methyltetrahydro-2H-pyran-2-one (2-2-11),4,6-dihydroxy-3,8-dimethyl-2H-chromen-2-one(2-2-4),4-hydroxy-3-(1-hydroxyethyl)-5,7-dimethyl- benzofuran-2(3H)-one (2-2-5). Compounds2-2-4and2-2-5were novel compounds.Six compounds were isolated from extraction of liquid fermentation of Verticillium dahliae Kleb.. All of the compouds were the first time found in this strain. The compounds were indole-3-carboxaldehyde(2-3-1),4-hydroxybenzoic acid (2-3-2),1H-indazole (2-3-3),2-(2-hydroxy phenyl) acetic acid (2-3-4),2-(1H-indol-3-yl)acetic acid (2-3-5),4-methylbenzoic acid (2-3-6).The antitumor activity research using MTT method showed that compound2-1-10had the inhibitory activity to the Hela cell strain, while the IC50of this compound was higher than positive control cisplatin. Compounds2-1-7had the inhibitory activity to Hela cell strain after48h. The activity of compounds2-2-2and2-2-3increased with the time passed. The activity may be increased further by the means of structure modification. Compound2-2-7and2-2-8showed no activity.Antioxidant activity research showed that the positive control BHA had the significant inhibitory activity with the change of concentration and the maximum rate reached to82.6%. Compounds2-3-4,2-1-5,2-3-1showed the antioxidant activity. Among these compounds, compound2-3-4show the higher activity, of which the maximum rate was50.0%, still lower than BHA.
