Verticillium dahliae Kleb.is a facultative host plant pathogenic fungus that can cause many plant verticillium wilt,which is harmful to economic crops and is difficult to control and cause significant economic losses worldwide.In recent years,with the introduction of genomic data of Verticillium dahliae and the urgency of seeking the control method of cotton verticillium wilt.A large number of potential pathogenic genes have been studied by scientists and researchers.In this paper.a chitin synthase gene VdCHS7(VDAG-00419)containing the actin head domain(MMD),which may be related to the pathogenicity of Verticillium dahliae.The homologous recombinant DNA fragment of VdCHS7 was ligated by Agrohacterium tumefaciens-mediated fungi(ATMT)by linking the hygromycin resistance gene between the upstream and downstream DNA fragments of VdCHS7 using the gene knockout method based on the principle of homologous recombination Method)was transferred into wild-type Verticillium dahliae V592,and the wild-type VdCHS7 gene was replaced by the hygromycin resistance gene by recombination of the homologous region gene fragment,thereby obtaining knockout mutant strain ?VdCHS7.V592 and mutant strain?VdCHS7 were inoculated in wild-type Arabidopsis thaliana,and the knock-out virulence was decreased.The functional complementary model of knockout strain was obtained by the same technique.The pathogenicity of Arabidopsis thaliana was inoculated back Make up At the same time,the wild type strain V592 was used as the control,and the biological traits of' the obtained knockout strain and the complementary strain were analyzed.It was observed that there was no significant difference between the culture trait of the complementary strain and the knockout mutant strain and the wild type strain V592.In this study,the chitin synthase gene VdCHS7 was not related to the biological traits and was the causative agent of Verticillium dahliae by the construction of the knockout body and the biological traits and pathogenicity.The gene knockout of the knockout mutant was obtained.and the biological traits of the functional complementary strains were obtained.The biological traits of the functional complementary strains were consistent with the wild type of the knockout body.The wild type Arabidopsis was found to be pathogenic,and the gene VdCHS7 was reaffirmed.Dahliae pathogenicity,independent of the formation of biological traits.Through the study of the chondrocyte synthase gene VdCHS7 in the VII class myosin domain,the pathogen of V.Dahliae is well understood,which is helpful for the control of the plant verticillium wilt caused by the pathogen.
