Optical surface plasmon resonance (SPR) is an advanced detection technologywhich is mainly used for fast detection of the analytes by monitoring the changes ofrefractive index generated from biochemical reactions. Optical SPR technology hasextraordinary advantages during the detecting process, such as label-free, real timemonitoring biological molecules dynamic responses, nondestructive testing for the analytesand so on. SPR technology has vast potential for future development and been widely usedin drug screening, environmental monitoring, biomedicine and many other fields.Furthermore, it has been an effective approach in studying the binding kinetics of theinteraction of biomolecules.The optical SPR biosensor, photoelectric detection principle, spectrum modulationmethod, and an advanced optical SPR biosensor system designed by a laser line generatorwere introduced in this paper. The advanced optical SPR biosensor system was composed ofa micro flow cell, clamp, convenient replaceable biological chip device, photoelectricconversion circuit, linear array CCD and temperature control module. The Ethanol, HepatitisB antigen and antibody were respectively implemented in the experiments to validate theperformance of the advanced biosensor and to construct the resonance responses curve andSPR responses curve. Automatic temperature control system was designed by employing themicroprocessor PIC16F876A where a semiconductor temperature-control chip and theregulation and control of the temperature were performed via PID algorithm. Several kindsof dynamical models of biomolecular interactions established by the SPR biosensor wereanalyzed and demonstrated by experiments. These optical SPR response signals wereanalyzed by a polynomial fitting method, simultaneously, the resonance sites (pixels position)and kinetic parameters were obtained. The experimental results clearly indicate that theadvanced optical SPR biosensor has important significance in designing the portable opticalbiosensor, and the curve between ethanol concentrations and optical response values was established, and their correlation coefficients of-0.9352were obtained and test sensitivitywas987.35(pixels/volume fraction).
