Rhizoma Corydalis is a famous traditional Chinese herbal medicine that has been found to have the efficiency of promoting both blood circulation to remove blood stasis and circulation of qi to relieve pain. It has been used mainly for the treatment of stomachache as an analgesic and is also effective for the treatment of inflammatory, hemorheological and allergic diseases.In our systematical study on the chemical constituents of Corydalis yanhusuo W. T. Wang,20 compounds were obtained from the 60% ethanol extract of the dried rhizome of this plant through various chromatographic techniques. Their structures were elucidated by physico-chemical property and spectroscopic analysis. The compounds were determined as:l-tetrahydrocoptisine (1), d-corydaline (2), tetrahydroberberine (3), yuanhunine (4),l-tetrahydropalmatine (5), l-tetrahydrocolumbamine (6), dehydrocorydaline (7), dehydroyuanhunine (8), 13-methylpalmatrubine (9), dehydrocorybulbine (10), palmatine (11), berberine (12), 8-oxocoptisine (13), N-methylcanadine (14), N-methyltetrahydropalmatine (15), didehydroglaucine (16),7-formyldidehydroglaucine (17), glaucine (18), (+)-O-methylbulbocapnine (19), and nantenine (20), respectively. Among them, compound 8 is a new compound, compounds 9 and 17 are new natural products, compound 19 was obtained for the first time from Papaveraceae, compound 14 was obtained from this genus for the first time, and compounds 13 and 15 were firstly reported from Corydalis yanhusuo.The HPLC fingerprint spectrum of the 60% ethanol extract of Rhizoma Corydalis and the CHCl3 soluble part were established using HPLC-DAD.12 peaks in the fingerprint were designated, and the identified peak area was 73.0% and 86.0%, respectively.MTT or SRB method was used to test the cytotoxic activity of 16 compounds against three human tumor cell lines. Three quaternary protoberberine alkaloids (7,9 and 12) exhibited significant cytotoxic activity against MCF-7 cell line. compounds 7,12,18 and 20 exhibited significant cytotoxic activity against K562 cell line. Among them, compounds 7,9 and 18 also showed cytotoxicity against liver cell line (LO2), compound 12 selectively inhibited K562 and MCF-7 proliferation, compound 20 selectively inhibited K562 proliferation.The immunomodulatory activity of most compounds isolated from Rhizoma Corydalis were evaluated. The results showed that compounds 1 and 15 promote the proliferation of T lymphocytes in dose-dependent manner.Most of the compounds were assayed against NO secretion of endothelial cell line Eahy926. Among the 16 tested compounds, compounds 1、5、12 and 16 displayed promoting effects on different levels. The result suggested that the above mentioned compounds may correlate closely to the effect of total alkaloids from Rhizoma Corydalis on increasing coronary blood flow.The anti-myocardial hypoxia effects of most compounds isolated from Rhizoma Corydalis were tested using the model of rat cardiomyocyte hypoxia in vitro. Among the 16 tested compounds, compounds 1,2,5 and 12 showed protective effects on hypoxia damage to rat cardiac cell line H9c2.
