In the modern treatment, many drugs to be used, effect unstable, drug resistance, relapse easily. Surgical treatment may cause postoperative complications, high cost advantage and more dangerous. The effect of spa, biological feedback is uncertain. But Chinese medicine treats slow transit cons t i pat ion with the whole concept as a guide. It develop individual i zed treatment programs with the pat ient, the Chinese medicine herbs wi th good effect, low toxicity, low cost advantage, no drug resistance. However, the treatment of Chinese medicine is diversity, mobility and lack of unified and outcome of the standard, it makes promotional and operational bad. Therefore, looking for a kind of united dialectical and simple treatment way becomes an urgent problem to be solved. Related clinical studies have shown that Yang deficiency constipation and yin deficiency constipation are two major types of constipation. It commonly used herbs of the Hemp seed pill and Ferry Brew. In the basic research, most scholars believe that constipation is relevant with colon power, but in recent years, visceral sensitivity becomes the research focus. Lots of research found that irritable bowel syndrome is relevant with visceral sensitivity. But the study of STC and visceral sensitivity is less, whether there is relationship between them is the focus of our research. Commonly used in the treatment of constipation the Hemp seed pill and Ferry Brew and how affect visceral sensi t ivi ty is a problem worthy of study, while the two effective prescription how to improve constipation condition is also worth exploring.ObjectiveBy analyzing the different groups, stool characteristics of the normal group, the STC model group (rhubarb group), STC overlay the Yin group (the Yin group), STC superimposed Yang group (the yang group), Hemp seed pill group and Ferry Brew group. And its gastrointestinal transit time, colonic pain threshold,5-HT4transporter body, c-kit gene and pathological features. Analyzing the occurrence of visceral sensitivity in the relationship of the STC, Yin deficiency and Yang the difference between the deficiency and to explore the mechanism of Hemp seed pill and Ferry Brew, fried on improving constipation condition.Methods (1)Preparation of STC animal model60healthy male big SD were selected,10SD were randomized into normal group, others were model group First of all, STC rat model was prepared by intragastric administration, the qualified model was based-model, then they were randomly divided into five groups (n=10), that is, the base-odel group, Yang deficiency model group, Yin deficiency model group, Yang deficiency and Jichuanjian group, Yin deficiency and Maziren pill group. Yang deficiency model group and Yin deficiency model group model were manufactured by hydrocortisone and thyroxine(2)Drug intervention model All treatment groups were administrated drugs by intragastric administration after the models were prepared successfully. The normal group, Yang deficiency group and the Yin deficiency group was given the same amount of water(3)Detecting indicators At the end of the experiment, gastrointestinal motor function was judged by the proportion of wet and dry of stool, gastrointestinal transit time of stool, the colon visceral sensory funct ion was determined by pain perception threshold. In order to explore the model of colon visceral sensory-motor dysfunction and colon tissue enteric nervous system-Cajal interstitial cells-smooth network-related gene transcription level of expression RT-PCR was used to detect the mRNA expression of colon tissue c-kit,5-HT4transporter. Using HE staining to observe the changes of the biopsy characteristics.Results(1) Through literature study for treatment of constipation, we get prescription134. After the reorganization of the statistical analysis and traditional Chinese medicine, the hemp seed pill and Ferry Brew is commonly used prescriptions for the treatment of constipation and the effective and reliable.(2)Compared with Yang deficiency group and Yin deficiency group, visceral sensation threshold raised on the hemp seed pill group and ferry brew group was lower(P<.05). The normal group, Yang deficiency group and Yin deficiency group also rised compared with the rhubarb group (P<0.05) and no obvious difference with rhubarb group.(3)Compared with rhubarb group, there is statistical significance on the normal group, Yang deficiency group and Yin deficiency group in the time through the gastrointestinal tract (P<0.05); The hemp seed pill group and ferry brew group is difference with Yang deficiency group and Yin deficiency group (P<0.05), and no obvious difference with rhubarb group. The time through the gastrointestinal tract in Yin deficiency group was longer than the Yang deficiency group, but there was no significant difference. (4)Hemp seed pill and ferry brew can improve the moisture content. Compared with Yang deficiency group and Yin deficiency group, it had significant difference (P<0.05), while was not statistically significant compared with normal group. Rhubarb group, Yang deficiency group and Yin deficiency were also different with normal group (P<0.05). The content of water in Yin deficiency-group was less than the Yang deficiency group, but there was no significant difference.(5) Through pathological examination, it found that smooth muscle reduce and lymphocyte increase in the rhubarb group, compared with the normal group. Lymphocytes dense clusters and even invade the mucous layer, and smooth muscle cell body atrophysmooth muscle layer destruction, organization and reduce the structural disorder in the Yang group, Yin group and the rhubarb group, compared with the yang group, lymphocytes was significantly more, the structure was more disordered, and even smooth muscle disappear in the Yin group. The hemp seed pill and the ferry brew were able to reduce the infiltrat ion of lymphocytes, enhancing smooth muscle, and has a clear structure.(6) The gene express ion level of c-kit、5-HT transporter are improved compared with the model group in the hemp seed pill and Ferry Brew group, but not significant difference. Compared with normal group, there were significant (P<0.01). In c-kit, the expression of the yang deficiency group was lower than yin deficiency group. Hemp Seed Pill and Ferry Brew group can improve c-ki t, there were no deficiency. In5-HT transport body, Compared with normal group, there were not significant in Hemp Seed Pill group. The expression of the yang deficiency group was higher than yin deficiency group.Conclusion(l)The influence of the hemp seed pill and ferry brew treating constipation is obvious.(2)STC is related to visceral sensitivity.(3)The hemp seed pill and ferry brew may be through increasing the water content of the waste and moist the intestine and low down visceral sensitivity to change constipation condition.(4)The hemp seed pill and ferry brew can improve5-HT transport body and c-kit and colon pathomorphological.(5) The Yin deficiency constipation may be difference with the Yang deficiency constipation. |
