Thalassemia is a hereditary blood disease which is caused by inborn gene defect, and this can hinder the synthetic of hemoglobins. HbHdisease is a common type of the intermediate α-thalassemias, which is common in the south province of china, and there is even on effect method to cure it. professor WuZhikui firstly proposed the hypothesis that "kidney generate marrow, marrow generate blood" is the core theroy for curing thalassemias by chinese herbs. Using YiSuiShengXue Granul to treat thalassemias in guangxi province have been30years and gained good effects. we study the mechanism from gene mutation,globin expression, the structure and function of the blood cell etc, and so to prove the objectivity of the hypothesis.From the clinical test and the animal test, this study use29a-HbH disease patients and mice with blood defect,which is irradiated by60CO γ.Fom the Activity and expression of hematopoietic cell factor, we use related technologies such as real-time PCR、SDS-PAGE、and RIA etc to study the mechanism of α-HbH disease by YiSuiShengXue Granul,and gain the results like below:1. the main results1.1The clinical research results of29α-HbH disease patientsWe treated29α-HbH disease patients conforming to the inclusion criteria using YiSuiShengXue Granul for3month,at last there were25patients finished the test, and there were24patient had effection,1patient had no effection. The Hb、HbH of the patients had an obvious improvement before and after treatment (P<0.05or P<0.01). RBC、Ret、PLT had no obvious improvement (P>0.05).This study indicated that YiSuiShengXue Granul could improve the peripheral hemogram of the α-HbH disease patients, which was in accordance with the clinical resul ts.1.2the clinical results of29α-HbH disease patients1.2.1Through testing hematopoiet ic cell factors in blood serum of24effective patients,we found that:3months after YiSuiShengXue Granul treatment, the activity and expression about SCF and GM-CSF had no obvious promotion (P>0.05), IL-3had a big decline (P<0.05). Hematopoietic cell factors mRNA PCR electrophores is result revealed that SCF gone up and IL-3declined. We could see that regulating the activityand expression of SCF、GM-CSF、IL-3might be a curing machanism.1.2.2We tested ED the MDA、SOD、GSH-Px from15patients,and found that after3month by YiSuiShengXue Granul treatment, GSH-Px had a statistical mean (p<0.05). this indicated that YiSuiShengXue Granul could ease the Oxidative damage of the red blood cells.1.3the results of the mice with blood defect, which was irradiated by60Co γ1.3.1Hb、RBC、PLT in the mice Peripheral blood had an obvious decliment after60C0γ irradiation,and we divided YiSuiShengXue Granul into diferent levels, and all of them had improved the level of RBC、Hb、PLT (p<0.01)1.3.2Three group of YiSuiShengXue Granul could enhance the indicators about spleen and thymus gland of the mice.we could see that YiSuiShengXue Granul could promot the hematopoietic capacity1.3.3Groups of different YiSuiShengXue Granul level could promote the content of SCF、GM-CSF in the serum (p<0.05), and decline the level of IL-3.To study the machanism of YiSuiShengXue Granul in treating α-HbHdisease from the hematopoietic cell factors, the clinical and animal tests had the same results,so we concluded that YiSuiShengXue Granul could improve the Hematopoietic function through adjusting the activity and expression of the Hematopoietic cell factors.2. The Innovation Points of This StudyTreating α-HbH disease patients as the research object, our study combined the tradional chinese medicine theroy and the molecular mechanism, and studided the molecular of treating a-HbH disease by YiSuiShengXue Granul from the aspect of Hematopoietic cell factors. From this study, we can give another proof for treating thalassemias by chinese herbs.
