PurposeTo observe the influence of Tong Xie Yao Fang to D-IBS model rats’colon tissue’s VIP、VIP1-R、AQP8. Disscuss the mechanism of Tong Xie Yao Fang. For the pain therapy to provide the basis for the mechanism.Materials and methods:30male Wistar rats were divided randomly into normal group, model group, traditional chinese drug group. Copy the rat model of diarrhea irritable bowel syndrome (D-IBS) by binding stress for18d,after D-IBS model was established suceesful, observed the general status including body mental status, skin color, weight and so on. traditional chinese drug group and antagonism group were taken medicine ig once a day in23.6g. kg-1.d-1for7days, determine the water content in rat feces. The method of reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry was used to determine the mRNA and protein expression of AQ P8、VIP and it’s receptor in colon tissue,Use the result to analyze the data.Results:1. D-IBS rats’ rate of weight:after the end of model, compare with the normal group, each group of rats’ growth rate had statistical significance (P<0.01); after the end of treatment,compare with the model group, traditional chinese drug group of rats’growth rate (?) statistical significance (P<0.01). D-IBS rats’fecal moisture percenta(?):after the end of model, compare with the normal group,each group of rats’ water content in feces had statistical significance(P<0.01); after th end of treatment, compare with the model group. traditional chinese drug group (?)f rats’ water content in feces had statistical significance (P<0.01).D-IBS rat’s the time of glass globule discard:after the end of model, compare with the normal group, each group of rats’ time of glass globule discard had significantly reduced, the differences had statistical significance (P<0.01); after the end of treatment, compare with the model group, traditional chinese drug group of rats’ time of glass globule discard had statistical significance (P<0.01).D-IBS rat’s histopathological examination of colon:Colon tissue of each rat was normal, Epithelial integrity, no congestion, edema, ulcers, inflammatory cell infiltration and no histopathological changes, showed a simple columnar epithelial mucosa, visible outer loose submucosa muscularis.2. Tong Xie Yao Fang for D-IBS rat’s colon tissue expression of VIP protein and VIP mRNA:compare with the normal group,each group’s rats’ colon protein of VIP and VIP mRNA expressed higher (P<0.01); compare with the model group, the chinese drug group’s protein of VIP and VIP mRNA expressed lower (P<0.01).3. Tong Xie Yao Fang for D-IBS rat’s colon tissue expression of VIP1-R protein and VIP1-R mRNA:compare with the normal group,each group’s rats’colon protein of VIP1-R and VIP1-R mRNA expressed higher (P<0.01); compare with the model group, the chinese drug group’s colon protein of VIP1-R and VIP1-R mRNA expressed lower (P<0.01).4. Tong Xie Yao Fang for D-IBS rat’s colon tissue expression of AQP8and AQP8mRNA:compare with the normal group,each group’s rats’ colon protein of AQP8and AQP8mRNA expressed lower (P<0.01); compare with the model group, the chinese drug group’s protein of AQP8and AQP8mRNA expressed higher (P<0.01)Conclusion:1. The weight of growth rate of D-IBS rat became lower、the water content in rat feces improved、the time of glass globule discard reducede. Tong Xie Yao Fang could reversed.2. The expression of the D-IBS rat model of colon tissue of VIP and VIP1-R protein and mRNA increased, Tong Xie Yao Fang could reduced the expression.4. The expression of the D-IBS rat model of colon tissue of AQP8protein and mRNA reduced, Tong Xie Yao Fang could improved the expression.
