Objective: To observe and discuss the influence of Ganoderma polysaccharides on theBDNF and Survivin content in the brain of rat with epilepsy caused by kainate acid (KA), andfurther study the epilepsy mechanism and anti-epilepsy mechanism of ganodermapolysaccharides.Methods:60male wister rats (weighing180~220g) were randomly divided into sixgroups:①the control group (n=10), in which rats received oral administration of the normalsaline and intraperitoneal injections.②Sham-operation group (n=10), in which rats wereinjected PBS phosphate buffer solution in right hippocampus and received oral administrationof the normal saline and intraperitoneal injections.③KA group (n=10), in which rats wereinjected Kainic acid into right hippocampus and received oral administration of the normalsaline and intraperitoneal injections. The behavior and EEG changes by deep electrode of therats were observed.④Ganoderma polysaccharides group one (n=10) in which rats wereinjected Kainic acid into right hippocampus and received intraperitoneally with Ganodermapolysaccharides (25mg/kg).⑤Ganoderma polysaccharides group two (n=10) in which ratswere injected Kainic acid into right hippocampus and received intraperitoneally withGanoderma polysaccharides (50mg/kg).⑥Ganoderma polysaccharides group three (n=10) inwhich were injected Kainic acid into right hippocampus and received intraperitoneally withGanod rats erma polysaccharides (100mg/kg)7days later,. Measure the content change ofBDNF and Survivin in cerebral hippocampus with immunohistochemical staining and LaserScanning Confocal Microscope.Results: All the rats of ganoderma polysaccharides group and epilepsy model groupachieved emblazed standard. In the KA group there was spike wave sharp wave andspike(sharp)-slow wave at the hippocampi. There was normal waves the sham-operation andnormal rats. Ganoderma polysaccharides group compared with epilepsy model group showt thatamplitude reduce decrease of frequency. In immunohistochemical staining experiment BDNFpositive cells of epilepsy model group increased obviously compared with the control group(P<0.05and ganoderma polysaccharides group increased obviously compared with epilepsymodel group (P<0.05). The positive cells of Survivin in hippocampus increased in the epileptic model group as compared with the control group (P<0.05); the positive cells of Survivin in theganoderma polysaccharides group was higher than that in the epileptic model group (P<0.05).Laser Scanning Confocal Microscope show BDNF fluorescence intensityof epilepsy modelgroup increased obviously compared with the control group (P<0.05) and ganodermapolysaccharides group increased obviously compared with epilepsy model group (P<0.05). Thefluorescence intensity of Survivin in hippocampus increased in the epileptic model group ascompared with the control group (P<0.05); the fluorescence intensity of Survivin in theganoderma polysaccharides group was higher than that in the epileptic model group (P<0.05).Conclusions: Both BDNF and Survivin may have relation with the starting anddeveloping of epilepsy caused by KA. Ganoderma polysaccharides could prevent the damagesof epilepsy to nerve cells by increasing BDNF and Survivin.
