Tumor is one of the main diseases which is harmful to human health and their life. Colon cancer is the second leading cause of cancer death in the western country and it takes fifth place in China. At the present, most of the chemical synthesis anti-tumor medicines will produce side effects to human body and influence the quality of patients’ life. So searching for the low toxicity and high curative anti-tumor activity ingredients form Chinese herbal medicine is the development direction of today’s anti-tumor medicines.This experiment wants to extract and separate polysaccharides from Ganoderma lucidum in order to classify them with the ultrafiltration method of the molecular quality.And then study the activity of different molecular components polysaccharide in vitro inhibiting proliferation of tumor cell. The study results are as follows:1With different volume of ethanol to concentrate and precipitate Ganoderma lucidum polysaccharide, getting three groups of them:50%group,66%group and80%groups’extraction rate respectively is2.59%、2.31%and3.08%. Comparing with Sevage method only and enzymatic with Sevage method, the latter one is the better method because having the higher protein removal rate and lower Ganoderma lucidum polysaccharides loss rate.2According to the membrane flux, searching for the optimal operating pressure, operating temperature and the concentration of Ganoderma lucidum polysaccharides of the different ultrafiltration membrane,the results is:Biomax500:0.08Mpa,40~45℃,1Omg/mL; Biomax300:0.06Mpa,20~30℃,1Omg/mL; Ultracel PL-100:0.08Mpa,40~45℃,10mg/mL; Ultracel PL-30:0.06Mpa,40~45℃,1Omg/mL.3Using ultrafiltration classification method,we get five group Ganoderma lucidum polysaccharides:GLPa≧1000000;600000≦GLPb≦1000000;200000≦GLPc≦600000;60000≦GLPd≦200000and GLPe≦60000.The content of polysaccharides of them is:GLPa,92.00%;GLPb,78.84%;GLPc,83.06%;GLPd,75.23%; GLPe,62.88%.60000~200000molecular weight group has the most propotion of all,47.19%;more than1000000molecular weight of polysaccharides’proportion is19.97%;200000~600000molecular weight of polysaccharides’proportion is13.90%;less than60000molecular weight of polysaccharides’proportion is9.50%;600 000~1000000molecular weight of polysaccharides’proportion is9.44%.4Screening from the five group of polysaccharides,200000~600000molecular weight of Ganoderma lucidum polysaccharides (GLPc) significantly inhibited the proliferation of colon cancer HCT-116. When adding GLPc mass concentration for625,1250,2500,5000and10000μg/mL, the inhibition rate were15.22%,34.36%,51.83%,77.43%and99.43%. There is a significant dose-response relationship between Ganoderm lucidum polysaccharides and inhibition rate.When the doses of Ganoderm lucidum polysaccharides adding to the highest,the HCT-116cell’s proliferation rate is less than1%.So Ganoderma lucidum polysaccharides GLPc have significant inhibiting tumor cell proliferation activity in vitro.
