Objective:This study through the detection MTA-2mRNA and protein inthe throat squamous cell carcinoma (LSCC) tissue of expression, analysis ofthe MTA-2expression and the clinical and pathological factors and therelationship between clinical significance, and also for the future MTA-2asthe early diagnosis of type of throat cancer-one of the index to lay thefoundation, and for the future of squamous cell carcinoma of the gene therapyprovides the basic theory basis.Subjects: the data of90patients with laryngostenosis squamous cellcarcinoma paraffin wax specimens, including40cases of matching material(original tumor and the corresponding lymph node metastases); Another20casesof cancer collection by10mm outside of the normal throat epithelial ascontrol. In order to verify the result of the immunohistochemical, wecollected30to throat squamous cell carcinoma and the normal throat cancerepithelial tissue of fresh specimens (corresponding mark) extract the mRNAand protein.By immunohistochemical method throat squamous cell carcinoma in tissuesamples from MTA-2of expression, and determined the MTA-2and the patient’sage and gender, and throat squamous cell carcinoma differentiation degree,PTNM stage, lymph node metastasis clinical pathological factors, therelations of the And detection MTA-2in the original tumor and thecorresponding lymph node metastases of expression differences.Use of RT-PCR and Western-blotting method for testing30throat squamouscell carcinoma and the normal throat cancer epithelial tissue samples in theMTA-2mRNA and protein expression, and statistical analysis verify they andclinical pathological factors of relevancy.SPSS for Windows13.0software analyzes the data. The Pearson ’sChi-Square test analysis MTA-2of the expression and throat squamous cell carcinoma patients with clinical pathological factors, the relations of theMTA-2in the original tumor and LN metastases expression of McNemar method,compares the RT-PCR and Western Blotting results using Independent-SamplesT test analysis. P<0.05is considered statistically significant.Results:(1)MTA-2in the throat squamous cell carcinoma and the throatcancer epithelial tissue of expression:90patients with laryngostenosissquamous cell carcinoma tissues, the MTA-2positive expression rate as highas64.4%(58/90), and they are all present in the cancer cells nuclear inside,and in20cases of the throat cancer corresponding epithelial tissue MTA-2for negative in expression.(2)MTA-2in the throat squamous cell carcinomaof the original tumor and the pairing of lymph node metastases of expression:MTA-2in the lymph node metastases to express the positive rate was85%(34/40),much higher than the pairing of62.5%of the original tumor.(3)MTA-2mRNAin the throat squamous cell carcinoma and the normal throat cancer epithelialtissue of the expressions: throat squamous cell carcinoma of the organizationfor0.99+/-0.08, the normal throat cancer epithelial tissue for0.38+/-0.14.(4)MTA-2protein in the throat of squamous cell carcinoma and the normalthroat cancer epithelial tissue of expression: throat squamous cell carcinomaof the organization for0.92+/-0.23, the normal throat cancer epithelialtissue for0.34+/-0.12.(5)MTA2mRNA and protein expression and throatsquamous cell carcinoma patients of the clinical and pathological featuresof relationship: MTA-2in low differentiation, high classification and havelymph node metastasis expression in the cases significantly higher, MTA-2ofthe expression and laryngeal cancer clinical PTNM staging was positivelyassociated with lymph node metastasis are related, and the cancer negativelycorrelated to the degree of differentiation, and gender differences with thepatient, age there is no significant relationship.Conclusions:(1) MTA-2of the expression and laryngeal cancer clinicalPTNM staging was positively associated with lymph node metastasis are related,and the cancer negatively correlated to the degree of differentiation, and gender differences with the patient, age there is no significant relationship.(2) the MTA-2mRNA and protein in the throat squamous cell carcinoma of theexpression level evidently higher than those of control of the normal throatepithelium of the organization, the difference between a statisticallysignificant.(3) MTA-2gene can promote throat malignant squamous cellcarcinoma of the proliferation and transfer, and can be used for throatsquamous cell carcinoma of the early diagnosis and an important marker andgene therapy targets.
