Objective:To investigate the influence of methyltransferase inhibitor5-aza-2’-deoxycytidine (5-Aza-CdR) on the protein expression status of RIZl gene in the human esophageal squamous carcinoma cell line TE13, and to study its influence on the cell proliferation.Methods:1.5-Aza-CdR was treated to the human esophageal squamous carcinoma cell line of TE13, Western blotting was used to investigate the protein expression status of RIZ1gene in the cells.Compare the RIZ1protein between the treated cells with the untreated cells, and during the cells suffer different time of the drug action.2.5-Aza-CdR was treated to the human esophageal squamous carcinoma cell line of TE13, FCM was used to study its influence on the proliferation of the cells. Compare the apoptosis rate between the treated cells with the untreated cells, and during the cells suffer different time of the drug action.Result:1. Compare with the corrosponding control group,the expression level of RIZ1protein in the48D and72D treatment groups was up-regulated after treated by5-Aza-CdR p<0.01).During the3treatment groups RIZ1protein expression level increased with the prolongation of the time of the drug action (p<0.01)2. Compare with the corrosponding control group,the apoptosis rate in all the3treatment groups was rised after treated by5-Aza-CdR (p<0.01).During the3treatment groups,the apoptosis rate rised with the prolongation of the time of the drug action (p<0.01)Conclusion:1、Specific phenolomethyl shift enzyme inhibitor5-Aza-CdR could up-regulate the protein expression level of the RIZ1gene in the he human esophageal squamous carcinoma cell line of TE13,further point out that promoter hypermethylation may be a contributory cause in the epigenetic silencing of RIZ1gene expression.2、Specific phenolomethyl shift enzyme inhibitor5-Aza-CdR could promote the apoptosis rate of the human esophageal squamous carcinoma cell line of TE13.Its role more clearly with the prolongation of the time of the drug action.This phenomenon indicate that promoter hypermethylation may be a contributory cause in the occurrence and development of partial human esophageal squamous cell carcinoma.
