Objective:To evaluate the male fertility and its Fl generation influence of early embryonic development of Sinomenine hydrochloride.Method:SD male rats were randomly divided into four groups.All animals are administrated by gavages for10weeks continuously, then delivery them into the same cage according to the male:female is1:2. Observe animals’ general situation.The pregnancy rats were dissected at the20th day after pregnancy. The mating rate and fertility index of rats were calculated. Record corpus luteum number, live birth number,stillbirth number, absorption fetal number,etc.And watch live embryo weight, appearance, internal organs and bone morphology, etc.Males were put to death after the mating,The testosterone levels of male rats serum and the glucose6-phosphate dehydrogenase levels of testes were determined.And use the HPLC to determinate the content of Sinomenine in serum, testis and epididymis tissue.Results:Each animal was in good condition during test.The rat’s mating rate and fertility index of each Sinomenine hydrochloride dose was no obvious abnormalities (P>0.05).Apparently unusual The female rat’s corpus luteum number,live birth rate, still birth rate and absorption rate of fetal also has not seen the obvious anomaly (P>0.05);Early embryo growth did not appearobvious anomaly (P>0.05).Compared with the blank group,there was no statistically significant difference in testosterone levels in serum and glucose6-phosphate dehydrogenase in the testis of Sinomenine each dose group(P>0.05).All the Sinomenine content in rat serum and testicular,epididymis tissue were inferior to the detection limit.Conclusion:Administrated Sinomenine hydrochloride for10weeks continuously at240mg/kg did not bring the apparent reproductive toxicity on the fertility and early embryonic development for SD healthy rats. According to body surface area, This dosage was equivalent to11times the quantity of clinical dose.So it could not cause obvious toxicity in male reproductive aspects by using Sinomenine hydrochloride in current clinical recommended doses.Administrated Sinomenine hydrochloride for10weeks continuously then stopped for a week,there was no cumulative effect in rat serum and reproductive tissues.
