Purpose:To compare the intervention mechanism of iodine、Haizao Yuhu Decoction in treating goiter caused by iodine deficiency from the perspective of anti-oxidative stress.Material and method:4-week-old Wistar rats were made animal model of iodine deficiency goiter.randomly divided into four groups:In addition to theNormal control group(NC) was fed normal diet, the other groups were fed a low iodine diet. NC group and Model control group(MC) group daily were fed with double-distilled water. Iodine excess group (IE) was fed with potassium iodate solution with the iodine concentration of1900μg/L. Haizao Yuhu Decoction group (HZ) group was daily fed by the same body surface area with human conversion of Haizao Yuhu Decoction. After giving treatment factor0week and8weeks, rats were killed. Rat serum GSH-Px、XOD and SOD activities, MDA and H202contents were determined by biochemical methods. Immunohistochemical method to detect thyroid4-HNE expression. PRDX5expression in rat thyroid and distribution were analyzed by Western blotting and immunohistochemical methods.Results:1、After treatment8weeks, the median of urinary iodine of model group was significantly lower than that in normal group, while IE group and HZ group the median of urinary iodine were significantly higher.2、After treatment8weeks, the model group TSH was significantly increased (P<0.001), TT3/TT4were significantly decreased (both P<0.001). IE group and HZ group TT4were significantly higher(bothP<0.05), TSH and TT3are no remarkable difference.3、After treatment8weeks, HZ group goiter recover better then IE group, the relative weight of thyroid of HZ group was lighter. Under the light microscope and electron microscope both observation of thyroid damaged lighter.4、After treatment8weeks, The serum SOD activity i in HZ group was significantly higher than the other groups (allP<0.05). the serum GSH-PX HZ was increased, but no significant difference-HZ group the expression of PRDX5in thyroid cells was significantly higher than those in the other groups. 5、After treatment8weeks, IE group serum H202content, MDA content, XOD activities and thyroid4-HNE positive were significantly higher than those in other groups (all P<0.05)Conclusion:1、HZ group goiter recovered completely. IE groups remained significant thyroid.2、IE group and HZ group were in a risk of hyperthyroidism, iodine in the treatment of goiter case by iodine deficincy lead to hyperthy roidism inevitably.3、Antioxidant capacity of the strongest for HZ group, antioxidant capacity decreased after iodine excess treatment4、Oxidative stress iodine excess group was highest, while HZ group oxidative stress level is lower.
