Objective:Taking TCM and acupuncture theory as the guide, to observe the regulation effects of electro-acupuncture on chronic fatigue rats’pain threshold,5-HT and P-EP, and explore the central regulation and control mechanism of pain-relieving by electro-acupuncture in CFS.Methods:Healthy SD rats were randomly divided into8groups:normal control group, model group. electro-acupuncture(EA) group, sham injection(SI) group, CRH group, CRH and electro-acupuncture(CRH-EA) group. CRH antagonist(CRH-A) group, CRH antagonist and electro-acupuncture(CRH-A-EA) group.Use the methods of compulsive cold water swimming in the daytime and chronic restraint at night to make fatigue model. Afterwards, observe SD rats’ complexion, and check rats in exhaustive swimming experiment. Morris water maze test, and hot plate pain measuring experiment.Then, inject in the lateral ventricles of SI group, CRH group, CRH-EA group, CRH-A group, CRH-A-EA group. With electro-acupuncture on shenshu and zusanli after injection, detect and analysis threshold of pain,5-HT and β-EP content in plasma,5-HT content in raphe nucleus.Results:①the results of hot plate pain measuring experiment:compared with normal control group’reaction time to pain, model group, SI group, CRH-A group,and CRH-A-EA group’shorter (P<0.05). Compared with model group’ reaction time to pain.EA group’, CRH-EA group longer (P<0.05)②the results of5-HT content in plasma:5-HT content of model group, CRH-A group and CRH-A-EA group higher than normal control group’content (P<0.05) CRH group and CRH-EA group lower than normal control group.③the results of P-EP content in plasma:model group, SI group,CRH-A group and CRH-A-EA group lower than normal control group. CRH group and CRH-EA group highter than normal control group.④the results of5-HT gray-scale value in raphe nucleu:compared with normal control group,model group,EA group, SI group.CRH-A group and CRH-A-EA group highter (P<0.05). CRH group and CRH-EA group lower than model group (P<0.05)Conclusions:after making model, chronic fatigue rats’ pain threshold reduced,5-HT content in plasma rised, β-EP content in plasma reduced, and5-HT content in raphe nucleu reduced, leading to sensitivity to pain. After electro-acupuncture treatment, chronic fatigue rats’5-HT content in plasma reduced, β-EP content in plasma raised, and5-HT content in raphe nucleu raised, leading to pain threshold raised. CRH is propitious to electro-acupuncture therapeutic action. Electro-acupuncture could modulate chronic fatigue rats’pain threshold. CRH is the key substance, while electro-acupuncture modulates chronic fatigue rats’ pain threshold.
