Objective:To take TCM and acupuncture theory as the guide, using the methods of ethology,chronobiology and molecular biology to explore the effect of electro-acupuncture's regulative action for spontaneous activity and hypothalamic CRH circadian rhythm of chronic fatigue syndrome rats.Methods:Health experimental SD rats were randomly divided into normal control group,model group and electro-acupuncture(EA) group. Using the methods of chronic restraint and cold water swimming to make fatigue model.EA group with electro-acupunture on shenshu and zusanli.To detect and analysis the ethology,spontanous activity and hypothalamic's CRH content of three groups after treatment.Results:①The results of ethology:rat tail suspension test,exhaustive swimming test,water maze effective zone residence time of normal control group compared with EA group, there were no significant statistical differences. Normal control group and EA group compared with model group, there were significant differences between two groups (P<0.05); water maze experiment time of normal control group compared with other two groups, there were significant differences (P<0.05), EA group compared with model group, there were significant differences (P<0.05).②the results of spontaneous activity:the peak phase,activity start time and end time of normal control group compared with model group, there were no significant statistical differences, normal control group and EA group compared with model group, there were significant differences between two groups (P<0.017); total activity of normal control group compared with model groups, there were significant differences (P<0.017), EA group compared with model group, there were no significant differences (P>0.017).③the results of hypothalamic's CRH content:there were obviously circadian rhythm both in normal control and EA group,model group's circadian rhythm disappeared; compared with normal control group and EA groups, the amplitude of model group has decreasing trend,but there were no significant differences; compared with normal control and EA groups, peak phase of model group has forward trend, but there were no significant differences. Conclusions:Chronic restraint and cold water swimming has been made rats body and psychological fatigue,chronic fatigue model was successful.The rat's body and psychological fatigue was obviously improved after electro-acupuncture treatment. Electro-acupuncture can regulated the spontaneous activity and hypothalamic's CRH circadian rhythm of chronic fatigue syndrome rats.
