Objectives:For the preclinical pharmacology research of FYN to explore t he mechanism of TCM compound,provides the basis theory for clinical researc h,screening effect safety and efficacy of comprehensive treatment of chronic pel vie inflammation disease of the newchinese herbal medicine.Methods:(1) Pharmacodynamics experiment:①To observe the anti-inflam matory action of FYN via oral administration and the effect of FYN on relievi ng pain, also observed the influence of engulfed function. We studied the the in fluence to the body blood rheology and microcirculation.②we used the phenol paste and mixed bacterial infection to cause rats CPID model.In addition to t he normal control group of10,the model rats were randomly divided into mod el group, FYN group, and FYN2.5,5, lOg original drugs per kg dose group, af ter20days of continuous administration, remove the two sides of the uterine, weighting compare swelling,then fixed with the formalin,embedded sections wit h paraffin,stained with HE, observe the pathological process of inflammation of the rat uterus with light microscopy and recorded and graded and scoring.Wei ghing and comparison the immune organ coefficient of the thymus and spleen of rats in each group (*g/100g weigh). Collected the rat’s abdominal aortic b1ood and determine the serum levels of T-SOD,MDA,IL-2,TNF-α,IL-6,Sig-A.Ob serve the changes in relevant indicators, Initially explore the mechanism of acti on of FYN to CPID.(2) Acute toxicity test:Given to KM mice, SD rat,res pectively, the maximum tolerated dose and the maximum quantity of administra tion of a single.Results:The test of pharmacodynamics indicate:FYN could restrain mouse ear tumefaction and rat foot tumefaction, Significantly against the acetic acid writhing test in mice caused by peripheral pain and formalin induced chronic p ain, Improve the phagocytic activity of chicken erythrocytes in mice. Significa ntly prolonged capillary clotting time as well as the microcirculation of mouse. Significantly inhibited the rat uterine swelling rate caused by phenol paste inf ection the CPID, Increased the index of the thymus and spleen. Elevated the r at serum levels of T-SOD and IL-2,reduce the serum level of the T-SOD and Sig-A of the CPID model infected by mixed bacterial (p<0.05or p<0.01).Th e pathology results also show that the FYN have better treatment to the CPID.(2) KM mice were single gavage FYN the maximum tolerated dose of93.2g (crude drug)·kg-1, equivalent to186times the recommended clinical daily oral dose, SD rats were give the largest single-dose administration the amount of crude drugper kg for68.8g, equivalent to138times of the clinical dose. D uring the continuous observation of two weeks after administration, no animal deaths and toxicity were found. It shows that the toxicity of the FYN’s single dose is small, it is safe in the clinical recommended dose.(3) after intragastr ically for six months of long-term toxicity tests, SD rats appeared abnormal su ch as general symptoms and signs, blood cytology, blood chemistry and tissue pathology and so on which caused by drugs, Recovery for2weeks, those abn omal were disappeared which indicate that the long term high-dose oral toxici ty of FYN is low, it can return to normal after withdrawal.Conclusion:FYN has a good role on anti-inflammatory、analgesic、regulat ion of the immune and the promotion of blood circulation microcirculation, it has a very good therapeutic effect for chronic pelvic inflammatory disease infe cted by chemical and pathogen, acute oral toxicity and long-term toxicity is lo w, which can be restored after stopping, it suggests that the clinical recommen ded dose or long-term is relatively safe, recommending for clinical treatment of chronic pelvic inflammatory disease and other gynecological diseases.
