Polygomum cuspidatum is the dried root or rhizome of Polygonum cuspidatum Sieb. et Zucc. The quality of polygonum cuspidatum is varied according to the different ecological environment. There were close relationship between the differences quality and micro-ecology of Polygonum cuspidatum. Researching on the correlation between endogeny fungi and plant metabolism, the causes of variance quality of Polygonum cuspidatum was revealed, and the scientific basis for the quality control of knotweed was provided.The167strains of entophytic fungi were generally found in the roots and rhizomas of Polygonum cuspidatum from different habitats of Pi County, Dujiangyan, Bazhong, Songpan Conuty, and were identified into2orders,4families and27genera. The dominant species were Moniliales and Melanconiales from the total number. The dominant species were Moniliaceae and Dematiaceae at branch level. The dominant species were Aspergillus, Fusarium, Geotrichum and Gliocladium on the classification of the genus level.The103strains of endogeny fungi of Polygonum cuspidatum which were separated from rhizome of the four habitats of Dujiangyan, Pi County, Bazhong, and Songpan Conuty were studied on liquid fermentation; the secondary metabolites of endophytic fungi of Polygonum cuspidatum were detected by TLC and HPLC. The results:by cooperation of strains G21and G33mixture ferment, the ability of producing polydatin was improved obviously, the other7strains were also found; by cooperation of strains G6-land3-4-2mixture ferment, the ability of producing emodin was improved obviously, the other3strains were also found; There were simultaneous determination polydatin and emodin in the fermentation products of strains of G6-1, G24and G12-2. At last,11strains of endogeny fungi were screened, such as G24,12-2,G12-1,3-4-2,2-4-3,G21.G33,G38-2, and G13-1,G6-1and G1-1-2.Study on the induction conditions of callus of Polygonum cuspidatum, the results indicated that the petioles for the induction of callus of Polygonum cuspidatum was the best explant, and the best induction hormone was â…¡ MS+6-BA3.0mg/L+KT0.2 mg/L+2,4-D0.5mg/L. The condition proliferation of callus subculture of Polygonum cuspidatum were optimized by the Box-Benhnken design, the best leve of hormones was MS+6-BA1.9mg/L+KT0.4mg/L+2,4-D0.5mg/L.The endogeny fungi elicitor and suspended cells of Polygonum cuspidatum were mixture cultured,the content of polydatin and emodin of co-culture cell were detected by HPLC,The results:By the elicitor of strains G6-1,G21,G13-1,12-2,3-4-2and the suspended cells mixture cultured, this Show that these elicitor of strains may promote cells suspended in the synthesis of polydatinr,the ability that G6-1,G21producing polydatin was improved obviously;the polydatin was produced by the strains of G12-1,G33,G38-2,G1-1-2,but the contents of polydatin were inhibited by the elicitor, this show that these elicitor of strains may inhibit cells suspended in the synthesis of polydatinr,the ability that G12-1,G33inhibiting polydatin was improved obviously;the polydatin was not produced by the strains of3-4-2, but the contents of polydatin were improved by the elicitor;the polydatin was not produced by the strains of2-4-3,G24, but the ability of producing polydatin in cell was inhibited obviously, this show that these elicitor of strains may block the synthesis of polydatinr.By the elicitor of strains2-4-3,G1-1-2,G24and the suspended cells mixture cultured,this show that these elicitor of strains may promote cells suspended in the synthesis of emodin,the ability of2-4-3,G1-1-2producing emodin was improved obviously;the emodin was produced by the strains of G33,G13-1,G38-2,3-4-2, G12-2,G21,12-2,G6-l,but the contents of emodin were inhibited by the elicitor,this show that these elicitor of strains may inhibit cells suspended in the synthesis of emodin, the ability that G33,G13-1producing emodin was improved obviously.All the results indicate that there are correlations between endophytic fungi separated from Polygonum cuspidatum and plant metabolism.
