objective:Establishing the NOD mice model of nonbacterial chronic prostatitis to explore the most effective compatibility proportion and selection of emodin,polydatin and olibanum volatile oil in Polygonum Cuspidate and Olibanum via Orthogonal Design Method.Method:220 male NOD mice which only 6 weeks old are selected referring to Virginia E.Rivero's method and were randomly divided into 11 groups:blank control group,model group,positive control group and other 8 groups in accordance with L8(27)orthogonal table.Each group has 20 mice raised under the condition of SPF.1 mg/ml of SD rat prostate protein purification liquid with double immune immune adjuvant is used on NOD mice except blank control group in 0 and 15 days to set up chronic nonbacterial prostatitis model.In the 21th day after the first injection of antigen,each mouse was treated gastric perfusion with 0.5mL of corresponding drugs in the corresponding group.Among them,the orthogonal design group gave gastric perfusion in different ratios of the emodin(100mg/kg.d;200mg/kg.d),titrin(200mg/kg.d;400mg/kg.d),volatile oil of frankincense(200mg/kg.d;400mg/kg.d);the positive control group gave the same amount of celecoxib 20mg/(kg.D),and the blank control group gave the same amount of distilled water to the stomach.The mice were executed by cervical kill after 14 days gavage administration.The prostatic tissue of mice was dissected and detected by ELISA method to survey the change of TNF-??IL-1??IL-6?IL-10 HE staining was used to study the pathological changes happened in prostate tissue.The evaluation indexes of prostate histopathological,TNF-?,IL-1 and IL-6?IL-10were evaluated to explore the most effective compatibility proportion and selection of emodin,polydatin and olibanum volatile oil in Polygonum Cuspidate and Olibanum via Orthogonal Design Method,which help to discuss the dose-effect relationship of Chinese herbal extracts'effect on NOD mice model of chronic non-bacterial prostatitis.Result:1.Histopathological observation showed that the prostate tissue in the model group accords with pathological changes of chronic inflammation.The glandular volume of prostate were adhered to adjacent tissue which is dark red or brown and slightly hard.Microscope shows that lymphocytes and monocytes infiltrated;interstitial edema and fibrous tissue hyperplasia are found;obviously along with prostatic epithelial damage,atrophy and exfoliation and prostatic wal hyperplasia or destruction.Statistical analysis was performed on the classification of prostate pathological sections,showing statistical significance(P<0.01)of the pathological differences between the normal mice and the model group(P<0.01),which indicates the success of the model.2.Impact evaluation of polydatin and olibanum on chronic non-bacterial prostatitis mice shows that the levels of TNF-??IL-1??IL-6?IL-10 in the prostate homogenation of mice had a certain difference.Compared with the normal group,level of TNF-??IL-1??IL-6?IL-10 in the prostate tissues of the model group was higher(P<0.01);level of TNF-??IL-1??IL-6?IL-10in the prostate tissues of the positive control group was lower than the model group(P<0.01);A more significant reduction appeared in 8 groups of orthogonal design,the difference was statistically significant(P<0.01);At the same time,the prostate tissues of mice showed obvious pathological changes.3.The results of orthogonal experiment:according to the evaluation indexes of prostate tissue pathological grade,TNF-?,IL-1?,IL-6,?IL-10 etc.,the efficacy of Chinese herbal extracts was evaluated comprehensively.The range of the comprehensive score showed that the effects of each factor from strong to weak were polydatin>emodin>olibanum volatile oil.The results of variance analysis showed that the effect of emodin on the curative effect was very significant,the effect of the emodin and polydatin on the effect was significant.The results of directly comprehensive analysis and variance analysis discovered that the optimal proportion of emodin,polydatin and olibanum volatile oil was 100mg/(kg·d),400mg/(kg·d),and 400mg/(kg·d).Conclusion:1.TNF-??IL-1??IL-6?IL-10's rise in prostate tissue of chronic nonbacterial prostatitis model reveals cytokines involved in the process of chronic nonbacterial prostatitis,through adjustment,the prostate tissue inflammation reaction can be alleviated or eliminated consequently relieve pain.2.Emodin,polydatin and olibanum volatile oil in Chinese medicine are effective on alleviating the inflammation of chronic non-bacterial prostatitis.From the overall and molecular level of the prostate tissue in mice,molecular herbal extracts and the treatment is expounded the action mechanism,confirming the efficacy authenticity of the treatment on chronic nonbacterial prostatitis.3.In addition,based on the orthogonal design of optimal proportion on emodin,polydatin and olibanum volatile oil,through comprehensive analysis of various indexes,the optimal matching was initially identified,which provides powerful basis for clinical use of this method.This study is innovative with clinical application significance,also provides a theoretical basis for the clinical application of new drug.
