Objective:To study the effect of substance P and its receptor antagonist onatherosclerosis in rats.Methods:36male Wistar rats were randomly divided into three groups, namely group A(control group), group B (substance P in the intervention group), Group C (substanceP+NK1R antagonist intervention group). The rats were treated with high fat diet andintraperitoneal injection of vitamin D3for building the atherosclerotic animal models.After12weeks, Two rats were killed from each group for pathological observationand lipids detection. Group A were continue fed with the high fat diet, and group Bwere given substance P on the basis of high fat diet, and group C were givensubstance P+NK1R antagonist (L-703,606) on the basis of high fat diet. On the14thweeks, all rats were sacrificed for blood biochemical character, lipid measurementand the serum hs-CRP or VEGF detection. The aortic tissue sections were stainedwith HE staining, toluidine blue staining and try immunohistochemistry.Results:1. The Pathological observation and lipid testing confirmed the success inestablishment of atherosclerosis model.2. The serum lipids, such as total cholesterol, triglycerides, LDL and high densitylipoprotein was no significant difference (p>0.05) in the three groups rats.3. The pathological changes of aortic atherosclerosis in the group B were moreseverer than the group A and C, while the group C were more serious than group A.4. The distribution of mast cells in the aortic atherosclerosis and degranulation of MCnumber of group A, group C, group B was gradually increased, group B than groupC (p <0.05), Group C than in group A (p <0.05).5. The level of serum hs-CRP in Group B was higher than that in group A (p<0.01)and group C (p<0.05). The level of serum hs-CRP in group C was higher thangroup A (p <0.05); The VEGF levels in group B were significantly higher than that in group A and group C (p <0.01), while group C than in group A (p <0.05).6. The correlation analysis of the serum hs-CRP and VEGF levels were shown with apositive correlation.Conclusions:1. Substance P promotes the accumulation of the mast cells in the lesion ofatherosclerosis.2. Substance P activates the mast cells, promotes atherosclerosis progress and may bereduces the plaque stability.
