Eczema Nano Cream Alternative Test Of Acute Toxicity In Vitro

Objective:Using Human KC Department of HaCaT cell culture in vitro,four methyl thiazolyl tetrazolium salt colorimetric (MTT)method was used to detect cell proliferation activity,to get an absorbance value (OD value), calculated cell inhibitory the rate of IC50according to the absorbance value to detect the acute toxicity of nano eczema cremor.Methods:KC Department of HaCaT in vitro subculture was purchased from the Central Laboratory of Xiangya. Nano eczema cremor was diluted into six concentration groups, and sodium dodecyl sulfate-positive control group, negative control group and blank control group,a total of9groups, respectively acting on the cells in each group after a certain time, detect cell proliferation activity by MTT method,obtain a group of absorbance value (OD value) and repeat the experiment for3times, calculated the cell inhibition rate according to the absorbance values, the cell inhibition rate of concentration logarithmic mapping, using SPSS16.0program by mapping method to calculate IC50.Results:MTT results showed that:according to the absorbance is eczema nano cream6concentration group from low to high cell inhibition rates were86.017%,85.317%,83.94%,84.14%,87.584%,90.767%,the inhibition rate of concentration logarithmic mapping, by mapping using SPSS16.0: IC50calculated for6.459mg/ml, IC50≥1.5mg/ml, showing that the nano eczema cremor minimal cytotoxicity or non-toxic.Conclusion:Using MTT method to detect the impact of nano eczema cremor on KC Department of HaCaT cell proliferation,The final result was: nano eczema cremor’s IC50is6.459mg/ml, the IC50≥1.5mg/ml. This demonstrats that nano eczema cremor minimal toxicity or non-toxic, nano eczema cream percutaneous absorption and short-term application toxicity produced is free or very small.
