Background: Eczema is caused by a variety of internal and external causes.It's an inflammatory change in the skin that tends to exudate,inflammation is more confined to the epidermis and the dermis shallow.The clinical manifestations of skin lesions are diverse,acute lesion is given priority to with exudation,erythema,papules,chronic skin lesions is given priority to with limited infiltrating hypertrophy,lichenification,it has urticant acuteness mostly,easy relapse.In recent years,a large number of studies have shown that although genetic factors,allergens and immune disorders are the basis of the pathogenesis,the infection of bacteria or the colonization bacteria is considered to be an important reason for the aggravation or recurrent of eczema.Recent studies have shown that the rate of colonization bacteria on the skin of eczema patients is significantly higher than that of normal people,and the severity of skin lesions is positively correlated with the number of microbial colonization.The use of a large number of antibiotics has caused the resistance of many bacteria on the surface of the skin,making eczema patients relapse and difficult to cure.It is necessary to find an antibacterial agent with good biocompatibility and high antibacterial activity.New nano material,nano enzyme is modified by specific substances,It has the ability to regulate ROS free radicals,can efficiently kill many gram-positive and gram-negative bacteria,and intractable biofilms,common nano enzymes aren ano-Iron oxide,nano-cerium oxide,nano-graphene nitrogen carbide,nano-gold and so on,they show very good antibacterial effect on escherichia coli,staphylococc us aureus,pseudomonas aeruginosa and streptococcus mutans and so on.Whether this nano material applications in infectious eczema have the same highly effective and safe antibacterial activity,according to the idea,this article aims to statistical analysis of clinical nummular eczemathe skin infection and research the effect and mechanism of nano-iron sulfide on murine infected eczema.Objective To research the positive rate and species of transplanting bacteria of skin lesions in patients with nummular eczema and the effect and mechanism of nano-iron sulfide on murine infected eczema.Methods 30 cases of skin lesions and non-skin lesions of patients with nummular eczema were collected for bacterial culture and identification,and statistical results;Mouse were sensitized by 2,4-dinitrochlorobenzene(DNCB)and innoculated with Staphylococcus aureus to establish a murine infectious eczema.Observe the improvement of n Fe S on mouse skin lesions;Count the viable bacteria in mouse skin homogenate liquid cultivation;Observe the histopathological changes of skin lesions by HE staining,and the expression levels of IL-10 and IFN-? serum were measured by ELISA.Results The positive rate of bacterial culture in skin lesions of eczema was 86.7%,among which the rate of staphylococcus aureus was the highest(63.3%).The detection rate of bacteria in skin lesions was significantly different from that in non-skin lesions(P<0.001);The mouse treated with DNCB and innoculated with staphylococcus aureus showed obvious erythema,papule,blister,accompanied by yellow scab.It shows that the infected eczema mouse model is established successfully.After treated with n Fe S,the skin lesions were improved markedly with decreased EASI scores.The number of bacteria of skin homogenate liquid cultivation decreased significantly(compared with the model group,P<0.01).Histopat hological analyses: in model group,hyperkeratosis and parakeratosis and acanth osis,and intercellular edema were observed in the epidermis,inflammatory cells infiltration in the superficial dermis,skin thickening and inflammatory cells infiltr ation were less in n Fe S group than that in model group;Serum inflammatory bi omarkers: compared with model group,the expression level of serum IL-10 in nFeS group decreased(P<0.01),while serum IFN-? increased significantly(P< 0.01).Conclusion the positive rate of skin lesion colonization was high in eczema patients,and staphylococcus aureus accounted for the majority.The n Fe S has significant bacteriostatic effect on Staphylococcus aureus and can ameliorate skin inflammation in mouse model of infectious eczema.The mechanism may be related to the regulation of the expression levels of serum inflammatory cytokines IL-10 and IFN-?.
