The paper consists two parts literature review and experimental research.First Section:Literature SummaryThere are two parts. The first one mainly introduces the research progress of the pathogenesis of depression, discussing from monoamine neurotransmitte、neuroendocrine、 immune system and genetic factor. The second one mainly introduces the progresses in the research on depression, discussing mainly from the Traditional Chinese Medicine treatment on depression in the past a few years Acupuncture treatment for depression principle and Clinical Research.Second Section:Experimental ResearchObjective:To observe the effect of praxeology indicators of chronic stress of depression model,the effect of electro-acupuncture on expression of different brain regions5-HT,DA,NE of chronic stress of depression model, discussing the mechanism of electro-acupuncture treatment of depression.Methods:Rats were randomly divided into six groups:controller (CON), model, binding,FLU,electro-acupuncture(EA),musical electro-acupuncture(MEA), using chronic unpredictable mild stress(CUMS) rats model, and collecting the samples21days later.(1) Using statistics to assess the effect of praxeology indicators of chronic stress of model;(2) Taking sample tissues from frontal lobe and hippocamp, then, using radioimmunoassay method, assessing the expression of Rat’s5-HT, NE, and DA towards electro-acupuncture.Results:1.21days after molding, using praxeology to assess the effects on rat’s activities, the results show, compared to controller group, the weight changes,, and the crossed squares of open field test of model group and binding group, the number of rearing of the rat have shown a decrease on average (P<0.01), intake of sugar water were significantly different (P<0.05); compare to model group, the crossed squares of open field test and the change of body weight are obviously higher in electro-acupuncture group and musical electro-acupuncture group (P<0.01); the change of body weight, intake of sugar water,the crossed squares of open field test,the number of rearing of the rats in PROZAC group (FLU) have shown significant differences (P<0.05); the measurements mentioned above between the electro-acupuncture group and musical electro-acupuncture group have no significant differences.(P>0.05) 2. Taking samples after successful molding rats, using radioimmunoassay to assess/detect the expression5-HT, DA, NE in different areas of rat’s brain. Specific Result As Following.(1) Hippocamp for the model group and binding group contains lower5-HT, DA and NE compared to controller group (P<0.01); compared to model group, hippocamp for PROZAC group (FLU) contains higher DA, NE, a significant difference (P<0.05).It also higher5-HT, a significant difference (P<0.01), and electro-acupuncture group shown a significant increment in NE (P<0.05); electro-acunpuncture and musical-electro-acunpuncture has no significant difference (P>0.05).(2) The content of5-HT, NE in frontral lobes of model group and binding group are obviously lower,comparing to controller group (P<0.01), and the content of DA in model group is lower than controller group (P<0.05); compared to model group, frontal lobes of rats in PROZAC group (FLU) has a significant increment of5-HT,DA,NE, shown a significant difference (P<0.05), while5-HT, NE in electro-acupuncture group are also increased significantly, shown a significant difference (P<0.05); there are no significant difference between in electro-acupuncture group and musical-electro-acupuncture group (P>0.05).Conclusion:1. The results of praxeology test shown that the rat’s exploration activities reduced, Weight loss,interest reduced after chronic unpredictable stress animal models.By using electro-acupuncture treatment,the chronic stress of rat’s depression behavior can be improved, this prove that the electro-acupuncture has antidepressant effect.2. Electro-acupuncture brings positive regulation to the expression of5-HT,DA, NE of chronic unpredictable stress animal models in different brain regions,electro-acupuncture can increase the level of5-HTs DA, NE,to achieve the effect of antidepressant. It’s proven that the mechanism of electro-acupuncture treatment on depression is to increase the level of central monoamine neurotransmitter of chronic unpredictable stress animal models.
