High performance liquid chromatography (HPLC) is one of the most importantmethods for separation and analysis in the subject of chemistry. Almost all the liquidorganic, polymer and biological samples can be analyzed by HPLC. It has beenwidely used in petroleum chemical industry, biochemistry, clinical medicine,foodhyginen, environmental monitoring, medicine industrial inspection and otherfields.The combination of HPLC and Chemical derivatization could satisfy therequest of the trace analysis of the most materials in nature. Developing newderivatization reagents for HPLC analysis is one of the active research areas forchromatography.Chapter one: The development history and basic theory about high performanceliquid chromatography and high performance liquid chromatography coupled withmass spectrometry were simply presented. The theory and application of chemicalderivatization with fluorescence derivatization were systematically introduced, citedwidely used derivative reagents in recent years.Chapter two: Pre-column derivatization methods for the sensitive determinationof fattyacids with10-ethyl-2-amino-acridone(EAA) as labeling reagents followed byHPLC-MS analysis have been developed. The optimization of derivatization andchromatographic separation conditions were evaluated.Then Put these methodsapplied to analyze tea.Chapter four: Pre-column derivatization methods for the determination of saturatedand unsaturated fatty acids with BAETS as labeling reagents followed byHPLC-MS analysis have been developed. The optimization of derivatization andchromatographic separation conditions were evaluated. The established methods wereapplied to the determination of acorn。Chapter Five: Pre-column derivatization methods for the sensitivedetermination of fatty acids with2-(12-benzo[b]acridin-5-(12H)-yl)-acetohydrazide(BAAH) as labeling reagents followed by HPLC-MS analysis have been developed.The optimization of derivatization and chromatographic separation conditions wereevaluated. The proposed method applied to fatty acid analysis from aconite to give themost satisfactory results.
