Glutamine synthase as a key enzyme plays a role in the central nervous system of the glutamic acid metabolism. This enzyme has some functions in maintaining the normal neural transmission, and repairing damaged nerves. Many cognitive disease are related with this enzyme mutation.Polyrhachis vicina Roger belongs to Polyrhachis, designated by the country’s health ministry, can be used for most food and medicine. This insect possesses some characteristics:grade-differentiation, complex social-behavior and highly complex nervous system. It is a good material for the study of insect growth development, neurological experimental research.The experiment and research results are as follows:1. Using nest’s RT-PCR,3’ and5’ RACE technology cloned the GInS gene cDNA from Polyrhachis vicina; the glutamine synthase gene’s length of the ant is1396bp. The longest open reading box for1110bp, code369amino acids,3’ the coding areas and5’ the length coding areas respectively for162bp and123bp,3’ the coding areas has a typically signal AATAA and ployA with19bp, which is the purpose gene, named pv-GInS, submitted to NCBI, get the serial number of the genes:JQ6790101.2. Using bioinformatics method to analyze and predict the GInS gene expression as well as the forecasted GInS protein. It’s molecular weight42.02kDa, isoelectric point pI5.70. For homology analysis showed that with other species especially insect glutamine synthase protein sequence similarity around75%. Structure domain analysis forecast the protein has glutamine synthase function areas,3. Using fluorescent real-time quantitative technology gain Pv-GInS’s mRNA level expression in the different development phase larvae and the different grade adultl. Experimental data shows that all of the test samples have Pv-GinS expression. In different development stages of larvae, one-instar larva, two-instar larva and pupa are expression of the high quantity, instead, eggs three-instar larva and four-instar larva of expression are low. In the different grade in adults, workers is the highest expression, the male is the more less than workers, and female ant at least.
