Objective:To observe the effect of Chinese medicine Qiang Gu Bao Formula through valuable-side interventions ovariectomized rat bone tissue functional changes in proteins,revealed the prevention and treatment of the menopause,osteoporosis sickness mechanism.Methods:1.Chinese medicine Qiang Gu Bao Formula suspension concentrated liquid preparation, WISTAR rat and removal of sub-ovariectomy osteoporosis,pine model,the conventional feeding and rearing of Chinese medicine intervention.2.In Sham-operated rats,ovariectomized rat model and the Chinese intervention in the rat model feur after 20 weeks of the compact bone,cancellous bone of the lumbar spine,such as tissue samples for protein extraction and study of sample preparation.3.With 2-D gel electrophoresis demonstrated two kinds of different groups of tissue samples proteome map.4.For each group the corresponding function of bone tissue proteome map to compare the differences.5.Applied Biosystems mass spectrometry and bioinformatics analysis of various quailtative differences in the function of bone tissue protein.6.Analysis and confirmed with postmenopausal osteoporosis-related incidence of bone tissue functional proteomics and traditional Chinese medicine intervention.Results:1.Postmenopausal rat model of osteoporosis Successful model:the Sham-operated group and model group,the average of the lumbar spine and femur bone mineral density have a very significant difference(P<0.01),model group and the Chinese intervention group aver-age lumbar spine and femur bone mineral density were significantly different(P<0.05),the Sham-operated group and the Chinese intervention group no significant difference(P>0.05).In accordance with the mean bone mineral density lower than 25%of the standard model group has two parts to achieve an average bone mineral density in osteoporosis standards,show that the model is successful.And Chinese parts of two intervention groups did not reach the bone mineral density, although the level of Sham-operated group,but the model group was significantly higher than that of Po-keung bone caused by ovariectomy decreased bone mineral density have a preventive role.2.2-D gel electrophoresis patterns obtained regarding the number of protein spots are as follows:femur in each group were 400,lumbar 600 are in each group.3.Comparison of femoral and lumbar spine specimen in the model group,the Sham-operated group and intervention group differences between Chinese protein expression,the difference between a total of 50 protein spots,25 femur specimens,25 lumbar spine specimens.Specimens of one of femur as a reference to the Sham-operated group,model group,six have reduced expression of proteins,4 protein up-regulated expression;Chinese intervention group down there is the expression of three proteins,three up-regulated expression protein;to the model group as a reference,there is medicine intervention group 5 up-regulated expression of protein,reduced expression of protein 4.Lumbar spine specimens to one of the Sham-operated reference group, model group,has 10 up-regulated expression protein;Chinese intervention group down there is the expression of five proteins,three up-regulated expression protein;as a reference to the model group,the intervention group of Chinese medicine have reduced the expression of protein 7.4.The difference expression 50 protein particle,uses MASCOT to search for the storehouse software to carry on the retrieval for the local database,is bigger than 61 by Protein Score to divide also C.I%>95%regard credible protein type,finally obtains 30 credible protein spots.5.After the biological mass spectrum technical appraisal,30 credible proteins hit the mark 5 are the unknown proteins.Finally confirmed that,The thighbone and the osteoporosis morbidity is possibly connected protein 6,respectively are:The lactoferrin light chain,membrane association protein A3,the enolase,ATP gather the enzyme,the acetyl coenzyme A reductases,the myo calcium protein;The lumbar vertebra and the osteoporosis morbidity is possibly connected protein 5,respectively are:The actin,the keratin,the enolase,ATP gather the enzyme,the myosin;The thighbone and the strong bone valuable curative effect is possibly connected protein 9, respectively are:The enolase,ATP gather the enzyme,the myo-calcium protein,the creatine activating enzyme isozyme,the phosphoglyceric acid change flavor the enzyme,the myosin,the lactoferrin light chain,the pyruvic acid activating enzyme isozyme,the crown protein;The lumbar vertebra and the strong bone valuable curative effect are possibly connected protein 8,respectively are:The carbonic anhydrase,the actin,the aB-crystal protein,3-phospho-glycerol aldehyde oxidase,the serum albumin,ATP gather the enzyme,the myosin,the enolase.Conclusions:1.From the femur bone density analysis of protein expression,we can see that light chain lactoferrin,annexin decreased and the expression of enolase,ATP synthase,acetyl-coenzyme A reductase,the expression of troponin,may be related to ovariectomized femoral bone density in osteoporosis-related happened;strong bone of the femoral osteoporosis Po intervention effect may be related to reduced enolase,ATP synthase,troponin,CK-MB,acetyl phosphoglycerate enzyme, the expression of myosin,and the increase in lactoferrin light chain,pyruvate kinase isoenzyme, protein expression crown.2.Cancellous bone from the lumbar proteomic expression analysis show that actin,keratin, enolase,ATP synthase,the expression of myosin may be related to ovarian lumbar cancellous bone after osteoporosis-related happened;Qiang Gu Bao Po on the lumbar spine osteoporosis intervention effect may be related to reduced carbonic anhydrase,actin,crystal protein,3-phosphoglycerate dehydrogenase,serum albumin,ATP synthase,myosin,the enolase expression.
