Purpose: To explore the effects of quercetin,vitamin E and amifostineon the damage of rats ovaries received cisplatin of intraperitoneal injection.Methods and Materials:50Adult female Sprague Dawley rats8-12weeks old were randomly divided into5groups: normal control group(A),quercetin-plus-cisplatin group(B), vitamin E-plus-cisplatin group(C),amifostine-plus-cisplatin group(D),and cisplatin group(E) groups. In themorning between8:00-9:00,the rats of group B and group C orally receivedquercetin(100mg/kg) and vitamin E(100mg/kg) respectively for a period of13days.The rats of the other three groups orally received equal vegetable oilfor period of13days. The rats of group D received intraperitoneal injectionof amifostine(200mg/kg) fllowed orally received vegetable oil at day1andday8.All the rats received intraperitoneal injection of cisplatin(6mg/kg) inhalf an hour after orally received vegetable oil at day1and day8,exceptgroup A. The rats were killed at day14.The Bodies and ovaries wereweighed.The avarin morphological and pathological were observed and theamounts of all levels follicles were measured.The superoxide dismutase in theovarian tissues were measured by WST-1way.The cytc protein in cytoplasmof the ovarian tissues, serum estradiol and follicle stimulating hormone weremeasured with ELISA kits.Then,statistical analysis.Results:1The bodyweight: There is no significantly difference of the bodyweightbetween the5group at the beginning of the study(P﹥0.05). At the end of thestudy: E vs C and D,B and C vs D bodyweight is significantly lower(P﹤0.05). B vs C and E bodayweight have no singnificantly difference(P﹥0.05).2The ovarian weight: B and E vs C and D,the ovarian weight issingnificantly lower(P﹤0.05).B vs E,C vs D ovarian weight have no singnificantly difference(P﹥0.05).3The follicles amounts:①primordial follicles amounts:E vs B and Cand D,B and C vs D,the primordial follicles amounts is singnificantly lower(P﹤0.05).B vs C,have no singnificantly difference(P﹥0.05).②Thegrowing follicles amounts:E vs B and C and D,B and C vs D,the primordialfollicles amounts is singnificantly lower(P﹤0.05).B vs C,have nosingnificantly difference(P﹥0.05).③T he mature follicles:There is nosingnificantly difference of mature follicles amounts in various groups(P﹥0.05).4The concentration of superoxide dismutase(SOD): B vs C and D andE,C and D vs E,the concentration of SOD is singnificantly higher(P﹤0.05).C vs D,have no singnificantly difference(P﹥0.05).5The concentration of cytochrome C(cytc):E vs B and C and D, B vsD,the concentration of cytc is singnificantly higher(P﹤0.05).C vs B and D,have no singnificantly difference(P﹥0.05).6The concentration of estradiol(E2):B and C and E vs D, B and E vs C,the concentration of E2is singnificantly lower(P﹤0.05).There is nosingnificantly difference between B and E(P﹥0.05).7The concentration of follicular stimulating hormone(FSH):B and C andE vs D,the concentration of FSH is singnificantly higher(P﹤0.05).There isno singnificantly difference between B and C and E(P﹥0.05).Conclusion:1Amifostine,vitamin E and quercetin could reduce cisplatin-induced ratsserum E2decrease and serum FSH increased.2Amifostine, vitamin E and quercetin could effectively reducecisplatin-induced rats the primordial follicles and the growing folliclesamounts rapid decrease.3The apoptotic protein cytc amount in the cytoplasm of ovarian tissuewould increased by cisplatin. Amifostine, vitamin E and quercetin can reduceit.4Amifostine, vitamin E and quercetin could significantly reduce cisplatin cause ovarian tissue SOD decrease.5Amifostine,vitamin E and quercetin could reduce cisplatin-induced ratbody weight and ovarian weight decreased.6Amifostine, vitamin E and quercetin can reduce cisplatin-induced ratovarian subcellular structural damage.7Amifostine can reduce cisplatin-induced rat ovarian damagesignificantly.
