Objective The study is to investigate effects of quercetin alone and quercetin combining cisplatin on proliferation and apoptosis of HepG2 cells and potential mechanism that quercetin reversed drug resistance of HepG2 cells to cisplatin .Methods The Growth inhibition effects of quercetin or quercetin combining cisplatin or cisplatin alone in HepG2 cells were detected by MTT; The apoptosis of HepG2 cells were detected by FCM assays; The changes of Bcl-2 and Bax were detected after dealt with quercetin or cisplatin or quercetin combining with cisplatin for 24,48,72 hours by immunohistochemical assays and image analysis system.Result Quercetin could inhibit the proliferation and promote the apoptosis of HepG2 cells which was associated with the concentration of quercetin (P<0.05); Under the same condition else ,growth inhibition and apoptosis of HepG2 cells were more obvious after dealt with quercetin combining cisplatin than dealt with cisplatin alone and the apoptosis of HepG2 cells were associated with the concentration of quercetin (P<0.05);The persistent increase of Bcl-2 and decrease of Bax and a continue increase of Bcl-2/Bax ratio were found in surviving HepG2 cells after dealt with cisplatin alone, but adverse thing was found after dealt with quercetin combining cisplatin .Conclusion Quercetin had effect of anticancer of hepatocellllar and could promote the chemiotherapy effects of cisplatin in HepG2 cells, which a probable mechanism was that quercetin reversed drug resistance of HepG2 cells to cisplatin through promoting increase of Bax and decrease of Bcl-2 in HepG2 cells.
