Objective: Bullous Pemphigoid(BP) is the most common autoimmune bullous disease under skin, characterized by extensive urticant blister,previously, it is considered that BP is Thl/Th2cell-mediated autoimmune diseases, and Th2cells as the advantages aim at its basement membrane adhesion molecules. However, in recent years, relationship betweenTh17cell type and autoimmune diseases has gradually be taken seriously. Because IL-17and IL-22are the important factors of Th17cells, studying the expression of the cytokines in patients with BP is helpful forrevealing the role of Th17cells in the pathogenic process of BP, andOpen up new ideas of diagnosis and treatment for BP.Methods:1Using immunohistochemical method to detect the expression of IL-17and IL-22in the skin lesions of41BP patients and25normal skin tissue.2Using ELISA method to detect the expression of IL-17and IL-22in the serum of27BP patients and25normal human.Results:1The immunohistochemical results1.1IL-17dyeing resultsThe expression of IL-17in cytoplasm and nucleus, and with skin full-thickness, leather shallow vascular endothelial cells and around the blood vessels of chronic inflammatory cells expressed in the skin tissue, no strong positive expression in normal skin tissues. In the41BP patients,19cases were strong positive,14cases were positive,5cases for weakly positive,3cases were negative; In the25cases of control group,no strong positive cases,2cases are positive,6cases are weak positive, 17cases were negative. Immunohistochemical staining results of IL-17in the lesion of BP patients are significantly higher than the control group, the statistics is significant difference(P<0.01).1.2IL-22dyeing resultsThe expression of IL-22is in cell plasma and mainly in skin epidermis of the organization, strong positive and positive expression in thenormal skin tissue is rare. In the41BP patients,13cases were strongpositive,14cases were positive,6cases for weakly positive,8caseswere negative; In the25cases of control group,2cases are strong positive cases,2cases are positive,4cases are weak positive,17caseswere negative. Immunohistochemical staining results of IL-22in the lesion of BP patients are significantly higher than the control group, the statistic-s is significant difference(P<0.01).1.3The correlation of IL-17and IL-22in the lesions of BPApplication of Spearman rank correlation analysis, r=0.213, P=0.181>0.01, IL-17in BP in patients with skin tissue was positively correlation with the expression of IL-22.2Using ELISA method detect IL-17and IL-22in the serum of BP patients and control group2.1The expression level of IL-17in serum (unit: pg/ml)Expression level of serum IL-17in BP group M (QR)=40.533(29.722), control group M (QR)=27.351(26.012), the expression level of BP cases is higher than the control group, the statistics is significant difference (P<0.01).2.2The expression level of IL-22in serum (unit: pg/ml)Expression level of serum IL-22in BP group M (QR)=39.231(30.000),control group M (QR)=23.776(16.349), the expression level of BP cases is higher than the control group, the statistics is significant difference (P<0.01).2.3The correlation of IL-17and IL-22in serum of BPApplication of two independent sample rank and inspection test the corr elation, r=0.886, P=0.000<0.01, BP patients serum IL-17was positively correlation with the expression of IL-22.Conclusion:1.The expression level of IL-17and IL-22in the skin lesions of BP is increased, it is explained that IL-17and IL-22can be involved inthe pathogenic process of BP.2.The expression level of IL-17and IL-22in the serum of BP is increased, it is explained that IL-17and IL-22can be involved in the pathogenic process of BP.3.The expression of IL-17was positively correlation with the expression of IL-22in BP, it is explained that IL-17and IL-22may play arole together in the pathogenic process of BP.4.The expression level of IL-17and IL-22in BP lesion and serumis increased, it is prompted that type Th17immune cells may be involved in the pathogenic process of BP.
